Outline of Research at the Start

申請者らは、天然多糖のβ1,3グルカンとDNAが3重らせん高分子複合体を形成することを初めて発見し、その基礎研究と核酸医薬のDDSへの応用展開を進めている。現在、CpGDNAやsiRNAを搭載した核酸医薬の臨床試験が進行中で、製剤化の容易さ、副作用の無さ、免疫細胞への高い選択性、超低用量が特徴である。本研究では、基本単位の分子構造の解析、新たに発見された量子化複合体の溶液物性の検討、多糖核酸複合体受容体であるヒトDectin1の発現細胞・組織の同定と役割解明、感染症・がん用の次世代高性能免疫刺激制御剤の開発、治療用細胞内ペプチドのDDSへの応用を目指している。

Outline of Annual Research Achievements

申請者らは、天然多糖のβ1,3グルカンとDNAが3重らせん高分子複合体を形成することを初めて発見し、その基礎研究と核酸医薬のDDSへの応用展開を進めている。現在、CpGDNAやsiRNAを搭載した核酸医薬の臨床試験が進行中である。本年度は、多糖核酸複合体受容体であるヒトDectin1の発現細胞・組織の同定と役割解明において大きな進展があった。今まで知られていないバリアントを発見すると同時に、新しい糖鎖修飾の位置も見出した。これらのバリアントの内DDSの標的となるバリアントを同定した。

Reason

多糖核酸複合体の基礎物性において、分子量が数万程度のβ1,3グルカンと複合体を作るときに、核酸が鋳型となって複合体が形成されることを見出した。これは、今まで考えていた多糖の3重らせんへの巻き戻りの過程にDNAが巻き込まれると言う、多糖鋳型説と正反対である。また、応用研究として進めている多糖核酸複合体としてのCpGの配列の最適化においても、機械学習を取り入れて進展があった。

Strategy for Future Research Activity

基礎物性としての構造解析には苦戦をしている。アルカリで複合体から核酸と多糖を分離することができることが分かったので、これを使って組成を決める。NMRは核酸のリンに構造異性があり、この分離ができないと不可能であると分かったので、R体もしくはS体からなる核酸を合成してその複合体のNMRを測定を試みる。溶液物性は論文化を目指す。応用研究の方は比較的順調に進んでおり、特許化を目指す。
