Outline of Research at the Start

ヒト造血幹細胞(HSC)を重度免疫不全マウスに移植した免疫系ヒト化マウスは、創薬における薬効や安全性の評価モデルとして広く認知されている。申請者は最近、NOG-W41マウスを作製し、放射線前処置なしでもヒトHSCの高生着性を有するヒト化マウスを開発した。本研究では、このNOG-W41マウスと各種次世代NOGマウスを組み合わせた次世代NOG-W41マウスを作製し、培養造血幹細胞移植と組み合わせた高汎用性ヒト化マウスモデルを開発する。

Outline of Annual Research Achievements

本研究は、新たなヒト化マウスの基盤系統として開発した、c-kit変異NOG-W41マウスへのヒトサイトカイン遺伝子導入による次世代型NOG-W41マウスの開発、およびこれらモデルへのヒト造血幹細胞移入による高生着性次世代NOGマウスの樹立を目指し、新たなヒト化マウスモデルの確立を進めている。
今年度は、前年度までに樹立し評価してきたNOG-W41-EXLマウスについて、汎用的多系列ヒト細胞分化モデルとして論文化した(Ito R et al. Stem Cell Res Ther. 2024 Jun 21;15(1))。また、NOG-G-CSF KIマウスにW41変異を導入したNOGW-G-CSF KIマウスを樹立し、これらへヒト造血幹細胞を移植したところ、従来のNOG-G-CSF KIマウスに比べて2倍以上のヒト好中球の生着性を確認した。本モデルを用い、LPS投与による生体内ヒト好中球遊走試験を今後実施する予定である。さらにヒト造血幹細胞を10倍程度増幅できる培養系を用い、これらへのGFP-Luciferase遺伝子を導入して移植後のヒト細胞の生着性を生体イメージングにより可視化できるシステムを確立した。これによりヒト細胞の局在を非侵襲的かつ経時的に追跡可能となり、ヒト化マウスにおける免疫応答や病態形成過程の解析への応用が期待できる。

Strategy for Future Research Activity

次世代ヒト化マウスの開発と評価をさらに進めるとともに、TCR遺伝子を導入した造血幹細胞を作製し、特異的な免疫応答を有するヒト化マウスモデルの開発を試みる。
