Outline of Research at the Start

がん細胞のARID1A発現低下がもたらす、がん微小環境における、がんと間質との相互作用における異常機構を検討することを目的とする。具体的には、胃癌、大腸癌でのARID1A発現低下による、がん間質の変化にどのような特徴があるか、を組織病理学的、人体病理学的に検討し、またin vitroでの、がん細胞ARID1A発現低下が、間質細胞とのNotch経路にもたらす変化の検討および、その制御方法の開発を検討する。

Outline of Annual Research Achievements

がん細胞のARID1A発現低下がもたらす間質の影響をin vitroで検討した。
ARID1A野生型、ARID1A変異発現低下型、人為的にARID1A発現を回復した大腸がん細胞株と共培養し、Exosomeによる未熟間葉系細胞への影響を検討した。
その結果、非接触状態ではみられないが、接触共培養系で、細胞間情報伝達システムに変化が生じることを網羅的遺伝子解析でみいだし、現在、検証中である。

ARID1A発現低下によるがん進行に、Exosomeを介したがん間質の変化がおこっていることが示唆された。
ARID1A発現低下によるがん進行抑制方法を、がん間質への働きかけの面から検討していく。
