Outline of Research at the Start

腫瘍組織では免疫細胞などの多彩な細胞が腫瘍の制御に関わっている。本研究では生体イメージング技術を用いて、腫瘍微小環境における免疫細胞の性状をリアルタイムに解析する。そして、がんの有無やその進展に伴う免疫細胞の変化を検出し、免疫細胞による効果的な悪性腫瘍の制御メカニズムの理解を目的とする。

Outline of Annual Research Achievements

本研究では最新の生体イメージング技術の1つである多光子励起イメージング技術を用いて、がん組織の腫瘍微小環境における免疫細胞の性状を、自家蛍光シグナルを手がかりとしてリアルタイムに解析し、さらにがん細胞の有無やその進展に伴う免疫細胞の生物学的な性状の変化を検出して、その背景因子をヒトサンプルも含めて詳細な検証を実施することで、免疫細胞による効果的な悪性腫瘍の制御メカニズムの解明を最終的な目標として研究を行う。上述した研究目的を達成するために、当該年度は研究計画に基づいて、本研究で観察対象としているマウスの実験計画整備と自家蛍光シグナルの検出系の確立、そしてこれに基づいた免疫細胞の同定を中心に検討した。

本研究で観察対象としている動物の実験計画を整えるとともに自家蛍光シグナルの検出系を確立でき、これに基づいた免疫細胞の同定を中心に検討を進めることができた。

Strategy for Future Research Activity

今後の推進方策としては、蛍光シグナルデータを経時的に取得し、その特徴量を定量的・統計学的に評価する手順を確立するとともに、スペクトル解析を併用して腫瘍組織特異的な蛍光シグナルの検出に努める。
