| Outline of Research at the Start |
NGS解析は網羅的解析である一方、コスト・解析に多大なエフォートを要するため、候補遺伝子スクリーニング目的でパイロットスタディとして実施する。その後の多数例解析には、高感度且つ変異特異的解析も可能なddPCRを用いる。2023-4年度にNGSを用いた候補遺伝子のスクリーニングを実施し、2025-2026年度にddPCRを用いた多数症例を解析し、有望なバイオマーカーを特定する。

乳房外パジェット病患者血清を用いて検討した。正常人と比較して、乳房外パジェット病患者では有意に血清cell free DNA濃度が増加しており、in situ群よりもinvesive群で有意な増加を認めた。また術後に血清cell free DNA濃度は減少した。さらに化学療法を行なった患者においては、血清cell free DNA濃度が病勢を反映していた。

乳房外パジェット病以外の皮膚悪性腫瘍患者での血清cell free DNA濃度の確認、さらに皮膚悪性腫瘍患者における個別マーカーの探索をすすめる。
