Outline of Research at the Start

先行研究の結果から、“HIF非依存性”、“長期低酸素刺激”、“癌周囲の非癌細胞と低酸素”が低酸素をめぐるダイナミックな癌形質の変化を解明するキーワードであると考えている。その一例として、長期低酸素刺激による大腸癌細胞の悪性形質の固定化が挙げられる。この変化は、HIFには依存しない経路でおこり、その形質が固定化されたものと考えられる。本研究では、大腸癌より樹立した初代培養細胞「オルガイド」を用いて、癌の多様性を保ったままでの検討をも同時に施行して行く予定である。時間軸と空間軸という観点から、低酸素のもたらすダイナミズムを統合的に解明する予定である。

Outline of Annual Research Achievements

本研究は、超長期にわたる低酸素環境が癌細胞の形質固定に与える影響を解明することを主な目的としている。生体内では癌組織内で癌細胞は極めて長期間にわたり低酸素環境に存在し、悪性形質を獲得していくことが予測される。具体的には、超長期的な低酸素状態における癌細胞の遺伝子発現の変化と、これが形質固定化にどのように寄与しているかを検討している。これまでの研究で、超長期低酸素下における広範囲なDNAメチル化変化が確認され、これが癌細胞の遺伝的安定性および形質固定に重要な役割を果たしていることが明らかになった。これらが低酸素環境に適応する過程で癌細胞に選択的な利益を提供している状況が示唆されているものと考えている。特定の遺伝子群のメチル化の変化をさらに詳細に解析し、現在、これらの遺伝子が具体的にどのような機能を担い、どのようにして癌細胞の生存と増殖に寄与しているのかを解析を進める予定である。
また、本研究では低酸素下での癌関連線維芽細胞(CAF)の挙動についても検討している。臨床サンプルからCAFの培養方法を確立した後、シングルセルRNAシーケンシング技術を用いて低酸素化に変化を受けるCAFの細胞ごとの遺伝子表現プロファイルを検討した。その結果、カルシウム代謝における変化が明らかになり、低酸素環境が腫瘍の進行や転移にどのように関与しているかの解明を進めていることが示された。このカルシウム代謝経路のシグナルがCAFの性質維持に重要な役割を果たしていることが示唆された。

Reason

本研究は、超長期にわたる低酸素環境が癌細胞の形質固定にどのように影響を与えるかを解明することを主目的としている。生体内での癌組織の状況を模倣し、癌細胞が長期間の低酸素環境下でどのように悪性形質を獲得し進化するかを検討するものである。当初の予定通りにおおむね研究が進捗している。

Strategy for Future Research Activity

これまでの検討では、超長期低酸素条件下でのDNAメチル化の広範囲な変化を確認し、これが癌細胞の遺伝的安定性および形質固定に重要な役割を果たしていることが示唆されている。特定の遺伝子群におけるメチル化の変化が低酸素環境に適応する過程で癌細胞に選択的なメリットを供与しているかどうかを、個別の遺伝子の発現や機能解析を進めることによって明らかにしていく予定である。
低酸素下での癌関連線維芽細胞(CAF)の挙動についても、臨床サンプルからCAFの培養方法を確立した後、シングルセルRNAシーケンシング技術を用いてCAFの細胞ごとの遺伝子表現プロファイルを詳細に検討し、カルシウム代謝経路における顕著な変化を確認した。これにより、低酸素環境が腫瘍の進行や転移にどのように関与しているかの解明を進めていく予定である。
