本研究は、希少難病である神経線維腫症２型の神経鞘腫に対して、新規薬物療法の開発を目指して、高度に糖鎖修飾された分子であるPTPRZを治療標的として、その詳細について基盤的な研究を行う。臨床検体を用いた検討ならびに、基礎実験を通じて、①糖タンパクPTPRzが神経鞘腫の腫瘍細胞増殖に関わるのか、②関わるとすれば、これを標的として増殖抑制が可能かどうかを、③糖鎖修飾の特徴との関係を含めて明らかにする計画である。

| Outline of Annual Research Achievements |
神経線維腫症２型患者で、神経鞘腫の切除を実施する症例を対象として、腫瘍ならびに髄液検体の採取・保存を行い、これを解析するため、本学の倫理委員会の手続きを進めた。これに基づいて、症例の登録、サンプルの収集を行った。さらに、これらの一部を用いて、PTPRZタンパクの評価を行った。一方で、PTPRZに対する抗体は、市販のものをふくめ数種類あるものの、PTPRZは、高度に糖鎖修飾されていること、また糖鎖修飾が異なる可能性もあり、特異的な抗体作出を進めた。

抗体については、概ね目処が立っており、来年度以降、進捗が見込まれる。さらに検体の収集を続けるため、本学以外の施設に協力を求めていく方針である。
