Outline of Research at the Start

加齢に伴い増加するRANKLや、歯周病によるLPSは、転写因子NF-κBを亢進する。NF-κBは骨吸収を促進し骨形成を抑制することで骨粗鬆症を重症化させる。我々が見出したmicroglia-healing peptides 1(MHP1)は、RANKL-RANK結合阻害と、LPSとCD14の結合阻害という二つの異なるNF-κB経路の選択的阻害という特徴を有することより、高齢者や歯周病を合併した重症骨粗鬆症に対しても有効性が期待される。本研究の目的は閉経後・炎症性骨粗鬆症モデル動物に対するMHP1の安全性と有効性を検証し、作用機序を解明して臨床応用を目指すことである。

Outline of Annual Research Achievements

本邦で約1300万人と推定される骨粗鬆症は医療費や介護力の点から喫緊の課題である。加齢に伴い増加するreceptor activator of nuclear factor kappa B ligand(RANKL)や、骨粗鬆症患者に高頻度で合併する歯周病によるLipopolysaccharide(LPS)は、転写因子nuclear factor kappa B(NF-κB)を亢進する。亢進したNF-κBは骨吸収を促進し骨形成を抑制することで骨粗鬆症を重症化させる。当グループが見出したmicroglia-healing peptides 1(MHP1) は、RANKLとRANKの結合阻害と、LPSとCD14の結合阻害という二つの異なるNF-κB経路の選択的阻害というユニークな特徴を有する。
当初の計画通り、2023年度は卵巣摘出による閉経後骨粗鬆症モデルマウスとLPS投与による炎症性骨粗鬆症モデルマウスの作成法を確立した。またこれらのモデル動物にMicroglia-Healing Peptides 1(MHP1)を腹腔内投与し、骨密度の低下が抑制されることを見出した。

Reason

当初の計画通り、2023年度は卵巣摘出による閉経後骨粗鬆症モデルマウスとLPS投与による炎症性骨粗鬆症モデルマウスの作成法を確立した。またこれらのモデル動物にMicroglia-Healing Peptides 1(MHP1)を腹腔内投与し、骨密度の低下が抑制されることを見出した。

Strategy for Future Research Activity

当初の計画通り、2024年度はMHP1の破骨細胞・骨芽細胞・骨細胞に対する効果をin vitroで検証する。具体的には上記の細胞にMHP1(1, 3, 10, 30, 100μg/mL; 先行研究より濃度を設定)を加え、WSTアッセイ(細胞増殖)、real-time PCR(分化・機能マーカー等の遺伝子発現)、ウエスタンブロット法による細胞内シグナル伝達経路、RNAシークエンス解析による網羅的遺伝子発現等の解析を行う。
