Outline of Research at the Start

大規模疾患ゲノム解析手法であるゲノムワイド関連解析(genome-wide association study; GWAS)はこれまで多数の疾患感受性遺伝子を同定してきた。一方で、従来のGWASは解析の対象がゲノム上の小規模な遺伝子多型に限定され、より大規模な塩基配列の個人差である構造多型は見逃されてきた。本研究では、ロングリードシーケンス、ショートリードシーケンス、一塩基多型マイクロアレイの3層の大容量ヒトゲノムデータを効果的に組み合わせた統合解析により、日本人集団のゲノムに含まれる構造多型を網羅的に同定し、疾患感受性への寄与を解明する。

Outline of Annual Research Achievements

大規模疾患ゲノム解析手法であるゲノムワイド関連解析(genome-wide association study; GWAS)はこれまで多数の疾患感受性遺伝子を同定してきた。一方で、従来のGWASは解析の対象がゲノム上の小規模な遺伝子多型に限定され、より大規模な塩基配列の個人差である構造多型は見逃されてきた。本研究は、ロングリードシーケンス、ショートリードシーケンス、一塩基多型マイクロアレイの3層の大容量ヒトゲノムデータを効果的に組み合わせた統合解析により、日本人集団のゲノムに含まれる構造多型の網羅的な同定と、その疾患への寄与の解明を目的とする。本年度の研究においては、ロングリードシーケンス、ショートリードシーケンス、一塩基多型マイクロアレイのそれぞれのヒトゲノムデータが検出可能とする構造多型の特徴を概観するため、各種構造多型解析ツールの実データへの適用と相互比較を実施した。この結果、各技術が得意とする構造多型の種類が判明し、効率的な探索に資するパイプライン構築の方針が定まった。また、臨床的に指摘できる原因を有さない不育症・習慣流産(unexplained recurrent pregnancy loss)を対象とした大規模ゲノムデータ(およそ2万6千人)に構造多型解析を適用することで、本疾患の感受性に寄与するpredicted loss-of-funcitonコピー数多型の同定に成功した。
