Outline of Research at the Start

オスマウスと交配後のメスマウスから着床前期胚を回収すると同時に、同一個体の子宮から子宮内膜をシート状に単離する。組織培養で広く用いられる気相液相界面培養系を用い、胚と子宮内膜の共培養を行い、トロホブラストの子宮内膜への接着・浸潤過程や、胚発生過程を試験管内で経時的に観察する。マウスを用いて堅牢な実験系を樹立した後に、種々の遺伝子調節システムを用いて着床・発生の分子学的制御機構を明らかにする。また、ヒト検体を用いた実験系の確立も目指し、将来の画期的な不妊治療の確立を目指す。

Outline of Final Research Achievements

We established a co-culture system by collecting embryos and endometrial tissue from mated female mice and applying an air-liquid interface culture method using PDMS devices. This system successfully recapitulated implantation and post-implantation embryonic development in vitro, faithfully reproducing attachment and invasion processes, developing embryonic structures, and COX-2 expression in the endometrium consistent with in vivo observations. Inhibition of uterine COX-2 retarded implantation and reduced AKT activity in the embryo. Introduction of a constitutively active form of AKT into the trophectoderm rescued this phenotype, suggesting the existence of a molecular pathway linking maternal COX-2 and embryonic AKT. This work has been accepted for publication in Nature Communications (in press).

Academic Significance and Societal Importance of the Research Achievements

近年日本における生殖補助医療(ART)の需要は高まり、出生児の約10人に1人がARTによる。受精技術の成功率は高いが、着床は依然として成功率が低く、ART成功の鍵となる。着床は子宮内で生じる複雑な高次生命現象であり、直接観察や操作が困難であるため、研究が進みにくい分野だった。今回、子宮の体外培養法と胚との共培養系を確立し、着床の過程を体外で忠実に再現することに成功した。これにより、これまで解明が困難だった着床調節機構の理解が進み、反復着床不全の病態解明や着床診断技術の開発、そして胚の着床機能を活性化することで着床率の向上や反復着床不全の治療を図る「着床補助技術」の開発に繋がることが期待される。
