Outline of Research at the Start

高解像度の空間的トランスクリプト―ム解析を通して、卵巣明細胞がん組織を構成する個々の細胞の特徴を明らかにする。この最新の解析手法により、がん細胞を網羅的に・一細胞レベルで・組織学的局在を保ちながら解析を行うことが可能であり、がん微小環境の解明につながる。そして、高解像度の空間的トランスクリプトーム解析の結果は、モデル細胞等による実験を通して検証し、新たな治療戦略確立に向けた研究開発を行う。

Outline of Final Research Achievements

Ovarian cancer comprises various histological subtypes, each with distinct molecular characteristics and clinical courses, necessitating subtype-specific analyses. Recent advances in spatial transcriptomics, which enable RNA expression analysis with spatial information, have facilitated more precise understanding of disease pathogenesis. In this study, we applied this technique to investigate malignant transformation in ovarian mature teratomas and identified KLF5 as a highly expressed gene in cancerous regions. Functional analysis using an established cell line revealed that KLF5 contributes to cell proliferation. In addition, in high-grade serous ovarian carcinoma, comparison between PARP inhibitor-sensitive and -resistant cases suggested the involvement of midkine signaling in drug resistance. Furthermore, RNA sequencing of gestational trophoblastic disease indicated that upregulation of XBP1 may contribute to the progression from hydatidiform mole to invasive mole.

本研究では、RNAの空間的な発現情報を取得できる新しい次世代シーケンス技術である空間的トランスクリプトーム解析を卵巣癌に適用し、腫瘍組織内の遺伝子発現の局在性を明らかにした。卵巣成熟奇形腫の悪性転化や高異型度漿液性癌におけるPARP阻害剤耐性に関与する分子を特定し、がん微小環境の理解が進んだ。これらの成果は、希少がんや治療抵抗性がんに対する新たな治療戦略の開発に貢献すると期待される。
