本研究は、Gli1-CreErt2/Rosa26-tdTomatoマウスにタモキシフェンな投与を行うことで、tdTomato蛍光を有するGli1陽性細胞を可視化する動物システムを採用して、骨組織におけるGli1陽性細胞の局在を解析する。Gli1陽性細胞はPTHによって活発に細胞増殖を行うか、また、Gli1陽性細胞が血管内皮細胞・血管平滑筋細胞に分化するのか、それとも、骨芽細胞へと分化するか、について、軟骨・骨移行部、骨幹端の骨梁、皮質骨骨内膜側、といった骨の各部位・領域に分けて検索する。

| Outline of Final Research Achievements |
In this study, we aimed to elucidate the effects of parathyroid hormone (PTH) on the proliferation, osteoblastic differentiation, and distribution of Gli1-expressing mesenchymal cells within bone tissue. Gli1-CreERT2; Rosa26-tdTomato mice were used, and the distribution of Gli1-positive cells was examined up to 14 days after administration of tamoxifen and human PTH [1-34]. In the metaphyseal chondro-osseous junction of the PTH-treated group, most Gli1-positive cells differentiated into ALP/PHOSPHO1-positive osteoblastic lineage cells. In contrast, in the trabecular bone area of the epiphysis, Gli1-positive cells differentiated not only into osteoblastic lineage cells but also into αSMA-positive vascular smooth muscle cells. However, in the diaphyseal region, Gli1-positive cells were not detected even after PTH treatment.

| Academic Significance and Societal Importance of the Research Achievements |
骨粗鬆症治療において、副甲状腺ホルモン(PTH)の遺伝子組換えペプチドであるテリパラチド(hPTH1-34)は、非常に優れた治療薬である。しかし、テリパラチドは骨幹端の海綿骨量を著しく増加させる一方で、骨幹部の皮質骨には多孔化を誘導することが知られており、海綿骨と皮質骨に対する作用が異なることが明らかにされている。今回の研究成果は、テリパラチドの海綿骨と皮質骨における作用の相違が、骨芽細胞の分化に依存する可能性を強く示した点で、学術的に大きな意義がある。また、本研究に基づき、テリパラチドの処方方法や骨粗鬆症患者における適応の最適化にもつながることが期待され、社会的意義も高いと考えられる。
