| Outline of Research at the Start |
未分化幹細胞に骨分化誘導剤を加えシャーレで2次元培養すると骨芽細胞分化が起きるが、細胞を凝集させ3次元培養すると骨細胞分化が促進されることを世界に先駆けて見出したが、そのメカニズムは未だに不明である。そこで本研究では、細胞核形状に注目し、「培養環境における細胞核形状が骨細胞・骨芽細胞分化の運命を決定する」という仮定の下、3次元及び2次元培養モデル内の核形状を定量評価する。また、組織器官形成に重要な役割を持つYAPの細胞核・ 細胞質移行と骨細胞分化の関わりも調べる。これらを通じて骨細胞分化機構が明らかになれば、試験管内で骨機能を模擬する骨オルガノイドモデルの構築に繋がる。

| Outline of Final Research Achievements |
In this study, we conducted mineralization induction within previously developed three-dimensional scaffold-free spheroids. Spheroids derived from human mesenchymal stem cells or mouse osteoblast-like cells were cultured long-term in osteogenic differentiation medium, and Alizarin Red S staining confirmed mineralization occurring from the inner regions of the spheroids. Additionally, to apply hydrostatic pressure to the spheroids, a hydrostatic pressurization system was developed. Application of 200 kPa static hydrostatic pressure significantly upregulated the expression of the Sclerostin (Sost) gene. Furthermore, using a glass plate compression test and atomic force microscopy (AFM), the mechanical properties of the spheroids such as global Young’s modulus, viscoelasticity, and local stiffness inside the spheroid were evaluated, revealing increased stiffness and heterogeneity associated with long-term culture.

本研究は、スフェロイド内での石灰化誘導および力学応答性の解明により、骨細胞分化機構の理解を深化させた。特に、静水圧刺激によるSost発現誘導や力学特性の定量評価は、骨恒常性のメカニズム解明と再生医療への応用に資する。これは将来の骨オルガノイド創成や創薬評価系への展開が期待される重要な知見である。
