これまでの研究で、頭皮細菌叢のポリアミン産生能力には非常に大きな個人差があり毛髪量との関連が示唆されたことから、我々は「プトレッシン高生産性の頭皮常在Staphylococcus epidermidisが頭皮にポリアミンを与えることで、宿主の発毛を促進している」という仮説を立てた。本研究では上記仮説を証明し、頭皮細菌叢の改善を通じた革新的な薄毛症治療法の確立のための基盤技術を開発することを目的とし、疫学調査、in vitro試験、動物実験を行う。現行の薄毛症の治療法で頭皮の共生細菌に着目したものは存在しないことから、本研究は挑戦的萌芽研究として、頭皮常在細菌と薄毛症との関連を解明する。

| Outline of Final Research Achievements |
In this study, we hypothesized that Staphylococcus epidermidis, which is highly productive of putrescine and is normally present on the scalp, promotes hair growth in the host by supplying polyamines to the scalp. We isolated 35 new strains of polyamine-high-producing S. epidermidis that accumulated 500 μM or more of putrescine in the culture supernatant from human scalp. In the first mouse experiment, the body hair area on the backs of mice in Group A (Difluoromethylornithine + polyamine-high-producing S. epidermidis HD25) showed a statistically significant trend compared to Group B (Difluoromethylornithine + PBS), but in the second mouse experiment, there was no significant difference in the body hair area on the backs of mice.

| Academic Significance and Societal Importance of the Research Achievements |
脱毛症の発症には主に男性ホルモンが関与することが知られており、治療法としては男性ホルモンの一種であるジヒドロテストステロンの合成酵素の阻害剤であるフィナステリドの処方等が推奨されているが、インポテンツ等の副反応も存在することから改善の余地が大きい。また現行の治療法は、宿主に直接作用するもので、頭皮の共生細菌に着目した治療法は存在しない。近年、皮膚常在細菌とアトピー皮膚炎をはじめとした様々な疾患との関連が続々と解明される中で、頭皮常在細菌と脱毛症との関連を解明し、脱毛症治療に効果的な頭皮のプロバイオティクスの社会実装を目指す本研究は、脱毛症治療薬の一大市場を形成する可能性を有する。
