Outline of Research at the Start

We will reconstruct full bacterial genomes from the gut metagenomes of the Ryukyuan termite species Nasutitermes takasagoensis and Odontotermes formosanus using long-read sequencing technology.

Outline of Annual Research Achievements

We sequenced the gut metagenome of Nasutitermes takasagoensis. We assembled long PacBio HiFi reads and obtained a highly contiguous assembly, including 14 full genomes of three dominant bacterial taxa in the gut of N. takasagoensis. These genomes inform us of the function of the main bacteria in the gut of termites. We also used these genomes to investigate the prevalence of horizontal gene transfers among bacteria residing inside the gut of termites. These results have been used to write one paper that we are about to submit for publication. We did not sequence the gut metagenome of Odontotermes formosanus but will sequence it during the second year of this project.

2: Research has progressed on the whole more than it was originally planned.

Reason

The project is progressing as initially planned. I originally planned to sequence the gut metagenomes of two species of termites from the Ryukyus, Nasutitermes takasagoensis and Odontotermes formosanus. We sequenced the gut metagenome of N. takasagoensis, assembled it and analyzed it. We also wrote one paper that we will submit during the next few months.

Strategy for Future Research Activity

I was planning to sequence the gut metagenomes of Nasutitermes takasagoensis and Odontotermes formosanus. While the work on N. takasagoensis has been performed and is about to be published, the work on O. formosanus has not been performed yet. I will sequence the gut metagenome of O. formosanus during FY24 and will analyze full bacterial genomes as we did for N. takasagoensis.
