Outline of Research at the Start

神経の発火が最大でも数百Hzという低さにもかかわらず、計算機を大きく上回る高速処理を脳が実行できるのは、脳の計算方式が超並列分散処理だからである。並列度を規定するのはシナプスの数であり、脳は膨大な数の神経細胞が約数千から1万のシナプス結合を有する超並列回路である。このような超並列情報処理の神経基盤の解明を可能とするイメージング法と、そのビッグデータ解析法を創出し、作動原理のモデリングを試みる。

Outline of Final Research Achievements

The aim of this project is to construct a two-axis, two-photon microscope system with synaptic resolution that can image the prefrontal cortex and other brain regions in mice simultaneously. First, we characterised the spatial resolution of the constructed microscope by measuring the fluorescence values and shapes of calibration microbeads. We then quantified the PSF of the XYZ axis from these values and confirmed that it had reached an acceptable level. Next, we aimed to achieve synchronised imaging of the two axes. As a result, we were able to achieve 3 ms misalignment in 1 min in real time and complete synchronisation of the two axes post hoc. In other words, we established imaging that enables the simultaneous high-resolution imaging of two brain regions, which is the main subject of this project.

Academic Significance and Societal Importance of the Research Achievements

本技術を使用することによって、2つの脳領域をシナプスレベルの解像度で観察および光操作し、その投射元を縦断的に観察することが可能になった。たとえば、双方向性に強い相互接続がある脳領域、たとえば、前頭野と扁桃体などが該当するが、各脳領域に、XCaMPsなどの色違いのCa2+インディケーターをそれぞれ発現させ、Head-fix課題遂行下での神経活動を同時計測したり、扁桃体を光遺伝学で操作した際の、前頭野でのセル・アンサンブルや行動の変化をオンラインで観察・操作することが可能になる。すなわち、脳の並列分散処理が、どのようにシナプス・細胞・回路に実装させているかを解明するオリジナルな手法と言える。
