がんの転移・浸潤・薬剤耐性などの悪性化には腫瘍微小環境が重要な役割を果たす。申請者は、グルタミンなどの非必須アミノ酸には、mTOR非依存的なゴルジ体を介した新規栄養センサーが存在する可能性を見出している。本研究は、これまで知られていないmTOR非依存的、且つがん細胞特異的な新規オルガネラ栄養センサーの探索を目的とし、がん特異的な新規栄養センサーを標的とした画期的な治療法の開発に繋げる挑戦的な研究としての意義を有する。本研究は、先天性アミノ酸代謝異常症などの小児難治性代謝疾患の新規メカニズム解明や治療法へと応用が広がる可能性も秘めている。

| Outline of Final Research Achievements |
This study aimed to explore and control novel cancer cell-specific nutrient sensors that are important for cancer cell survival and maintenance of stemness, and examined the following items. Establishment of gRNA screening system using organelle morphology as an indicator and identification of target genes: In this research component, (1) we established a system using a CRISPR gRNA library to sort cells with normal Golgi structure and cells with collapsed Golgi using imaging cytometry. (2) Using a cell collection device that can be operated under super-resolution microscopy, we were able to manually collect cells selectively under super-resolution microscopy using Golgi morphology as an indicator. In the future, we plan to identify target factors that maintain nutrient responsiveness using Golgi morphology as an indicator based on gRNA information from the collected cells.

| Academic Significance and Societal Importance of the Research Achievements |
アミノ酸経路はがん細胞のmTOR複合体を介した悪性化に重要だが、ラパマイシン等のmTOR阻害剤では根治に至らない。理由として、mTOR非依存的な栄養感知機構の存在が考えられる。本研究は申請者の低栄養培養系を用い、gRNAスクリーニング系とイメージングサイトメトリー、超解像イメージング細胞分取法を融合し、ゴルジ体を介した新規栄養センサーを同定する。mTOR非依存的栄養感知機構を標的とした次世代がん分子標的薬開発を目指す。オルガネラ栄養センサー発見による独創的アプローチで、がん治療法開発と先天性代謝異常疾患の病態解明への応用が期待される挑戦的研究である。
