Outline of Research at the Start

非アルコール性脂肪性肝炎 (NASH) は致死的疾患である肝硬変,肝癌へと進展する.肝臓と骨格筋にはクロストークが存在し(筋-肝連関),骨格筋の異常はNASH進展のリスクを増大させる.しかし,その詳細なメカニズムは未解明である.代表者らは,酸化ストレス防御の司令塔Nrf2を筋細胞でのみ発現するNrf2-筋レスキューマウスでは,筋-肝連関を介して肝病変が抑制されることを見出した.一方,細胞外小胞は細胞間の情報伝達を担う因子として知られている.本研究では,筋Nrf2がNASH を抑制する筋-肝連関メカニズムについて,Nrf2を介した細胞外小胞の内包基質の変化に着目して検証する.

Outline of Final Research Achievements

In present study, we analyzed the mechanism of NASH suppression by muscle Nrf2, focusing on muscle-derived extracellular vesicles (EVs). Experiment 1: EVs were collected from the medium of myotube cells (C2C12) treated with or without sulforaphane (Nrf2 activator). Proteomic analysis of the proteins contained in the EVs was performed, and compared with or without sulforaphane treatment. Sulforaphane-treated C2C12-derived EVs showed higher expression of antioxidative protein (Crz, Txnrd1, etc.) than vehicle-treated C2C12-derived EVs. Experiment 2: Sulforaphane-treated C2C12-derived EVs were added to the macrophage cells (RAW264.7) in which inflammatory condition was induced by LPS. The gene expression level in the cells was compared with that in the vehicle-treated cells. The expression level of inflammatory cytokines and chemokines was decreased by sulforaphane-treated EVs.

Academic Significance and Societal Importance of the Research Achievements

本研究では,Nrf2活性化に応じて筋細胞からEVsを介して抗酸化分子が分泌されることを明らかにした.また,このEVsは免疫細胞(マクロファージ)に作用し,抗炎症効果を発揮することが明らかとなった.筋細胞のNrf2はNAFLDをはじめとする炎症性疾患に対して防御的に機能することが示唆された.
