Outline of Research at the Start

Sprague-Dawleyラットを対象に,遺伝子を細胞内に運ぶアデノ随伴ウイルスベクターを用いて,運動神経のみにチャネルロドプシン遺伝子導入をする.導入されたチャネルロドプシンは,青色光の照射により活性化し運動神経を興奮させ,結果的に筋収縮を起こすことが可能となる.それによって生じる交感神経活動と血圧応答を測定し,従来の椎弓切除術を用いた方法と比較する.

Outline of Annual Research Achievements

本研究は,光遺伝学的手法を用いて,循環動態を制御する求心路を刺激せずに,運動神経のみ興奮させることで筋収縮を起こし,運動昇圧反射を評価する新規方法の確立に世界で初めて挑戦することを目的としている.
アデノ随伴ウイルス(AAV)6を生後2日齢のSprague-Dawleyラットの腓腹筋に筋肉注射し,4から6週間かけてその腓腹筋を支配する脛骨神経内の運動神経を逆行性に移動させ,神経特異的プロモーターを用いてチャネルロドプシンを運動神経のみに遺伝子導入した.この導入されたチャネルロドプシンは,研究者が制御する青色光照射により活性し運動神経を興奮させて結果的に筋収縮を起こすことが可能となった.電気刺激による最大単縮張力の約50%程度の単縮が光刺激により観察できた.しかし,強縮は多くの例で確認できず,光照射による強縮によって生じる交感神経活動と血圧応答を測定するには至らなかった.
