Outline of Research at the Start

本研究は、ゲノム解析・集団遺伝学理論からサツキマスの毎年の生息個体数を推定するとともに、最新の地球化学分析手法によって、サツキマス耳石のSr同位体比分析を行い、各個体の母河川や成長した場所を推定する。そして、結果を地図上で統合するアルゴリズムを開発し、長良川流域において、サツキマスのレジリエンス機能を維持するための流域ネットワークを可視化する。

Outline of Final Research Achievements

This study aimed to elucidate the habitat network supporting the resilience function of Oncorhynchus masou populations. The research developed an innovative framework combining otolith strontium isotope analysis with CKMR methods to visualize spatiotemporal dynamics of wild fish populations.
Using the Sr isotope map of the Nagara River that we constracted, we estimated birth locations of masu salmon individuals. Results revealed that while many masu salmon were produced in the uppermost reaches of the Nagara River, individuals from other watersheds were also present.
Furthermore, GT-seq analysis based on masu salmon genome sequencing estimated the effective population size (Ne) of the parent population of masu salmon that migrated upstream in the Nagara River. Combining these results with otolith isotope analysis quantitatively demonstrated that 92-552 masu salmon individuals inhabited each of four tributaries in the upper Nagara River and contributed to masu salmon reproduction.

Academic Significance and Societal Importance of the Research Achievements

日本では淡水・汽水域の魚類169種が絶滅危惧種に指定されており、効果的な保全対策が急務となっている。本研究で実施した耳石同位体分析とゲノム解析を組み合わせた手法は、従来困難であった魚類の詳細な生活史と個体数動態の把握を可能にすると考えられる。特に、ポートフォリオ効果の実証により、単一河川ではなく流域全体での保全の重要性が指摘された。この知見は、河川改修や環境変化に対する適応的管理戦略の策定、水産資源の持続的利用計画、生物多様性保全政策の根拠として活用可能である。また、本手法は他の魚種にも応用可能であり、日本の水域生態系保全と水産業の持続可能な発展に大きく貢献することが期待される。
