| Outline of Research at the Start |
細胞は、細胞外環境からの情報をもとに極性化が行われ、方向性を持った細胞機能を発揮すると考えられるが、極性化を制御する機構は未だ不明な点が多い。そこで本研究では、その分化過程において特徴的な極性化を示す細胞種に着目し、エナメル芽細胞、破骨細胞および骨芽細胞の3種類の細胞の極性化モデルを用いて網羅的遺伝子解析を行い、極性化に伴い誘導される因子のスクリーニングを行うことで、細胞極性化制御因子の同定を図ることを目的とする。

| Outline of Final Research Achievements |
Using the dental epithelial cell line M3H1, we established a cell polarity induction model using the basement membrane component laminin. We confirmed that cells induced to differentiate under laminin-coated conditions exhibited a morphological change, and the polarity marker Zo-1 localized to the apical side. Analysis of Zo-1 and cell adhesion-related gene knockout cell lines suggested that these genes are important for cell arrangement and the establishment of cell polarity. This study successfully developed a screening model capable of evaluating cell polarity, which is expected to contribute to the future elucidation of cell polarity control mechanisms.

| Academic Significance and Societal Importance of the Research Achievements |
本研究は、細胞極性制御機構の解明に向けた新たな実験モデルを確立した。これまで解析が難しかった上皮細胞の極性形成と分化の関連性を、簡便な培養系で詳細に調べることが可能になった。将来的には、このモデルを用いて細胞極性を制御する様々な因子を網羅的に探索することで、組織再生医療や、細胞の極性異常が関与するがんなどの疾患における新たな治療ターゲットの発見につながる可能性が考えられる。
