Outline of Research at the Start

不安や恐怖といった情動は、ラットなどの齧歯類からヒトを含む霊長類まで広く保存されていることが知られている。哺乳類は高い社会性を獲得する過程で、他個体とのコミュニケーションを通じて不安や恐怖を適切なレベルに制御するメカニズムを獲得したと考えられるが、このような制御メカニズムに関する理解は進んでいない。そこで本研究は、ラットにおいて不安や恐怖を惹起する警報フェロモンと、不安や恐怖を抑制する安寧フェロモンという、応募者が見出した2つの情動フェロモンの中枢作用メカニズムを解明する。そして両者を詳細に比較することで、不安や恐怖を状況に応じて適切なレベルに維持する情動制御メカニズムを包括的に理解する。

Outline of Annual Research Achievements

本年度に得られた主要な成果は以下の通りである。
警報フェロモンに関する研究
警報フェロモンのリガンドである4-メチルペンタナールとヘキサナールはそれぞれ鋤鼻系と主嗅覚系で受容されるため、これらの情報が「統合センター」にて統合された後に、分界条床核へと伝達されていくことが想定されている。この統合センターを同定するために検討を進めたところ、扁桃体内側核後腹部が統合センターであることが明らかになった。またその他にも、幾つかの系統のラットから警報フェロモンが放出された場合には、その存在を認識できないというように、警報フェロモン効果の系統差が存在することが明らかになった。
安寧フェロモンに関する研究
安寧フェロモン作用の神経メカニズムとして、前嗅核後部が重要な役割を担っていることが示唆されている。しかし前嗅核後部はこれまでほとんど解析が行われてこなかった神経核であるため、基盤となる解剖学的情報が非常に乏しい神経核であった。そこでトレーサーを用いて解析を行ったところ、前嗅核後部は扁桃体内側核後腹部や扁桃体基底外側複合体と解剖学的に接続していることが明らかになった。また安寧フェロモン作用においても、警報フェロモンの統合センターである扁桃体内側核後腹部が重要な役割を担っていることが示唆された。

Reason

警報フェロモンに関しては、鋤鼻系と主嗅覚系で受容した情報を統合する「統合センター」の候補となる領域を同定することができた。また並行して、鋤鼻上皮で受容されることが明らかになっているものの、その受容体が不明であった4-メチルペンタナールの受容体を同定することを目指しており、実際に絞り込みを開始することができた。そして安寧フェロモンに関しても、扁桃体内側核後腹部がその作用に関与していることを明らかにすることができた。さらに今後の研究進展に必須となる、前嗅核後部の解剖学的情報を数多く得ることができた。加えて安寧フェロモン分子に関する研究に関しても有力な候補物質を同定することができた。以上を勘案すると、研究計画はおおむね順調に進展していると考えられる。

Strategy for Future Research Activity

警報フェロモンに関しては、これまでの研究を継続し、発展させていく。すなわち、扁桃体内側核後腹部が1)候補領域と分界条床核が解剖学的に接続しており、2) 候補領域と分界条床核との接続が警報フェロモンを受容した際に活性化し、3) 候補領域と分界条床核との接続の活動を人工的に操作すると警報フェロモン作用が変容することを検討する予定である。また受容体同定に関しても現在の研究を進め、候補受容体を絞り込んでいく予定である。
安寧フェロモンに関してもこれまでの研究を継続し、発展させていく予定である。神経メカニズムに関する研究では、基盤となる解剖学的情報をさらに収集し、またフェロモン作用を生み出す神経化学物質を同定することを目指す。またフェロモン分子に関する研究では、候補物質がフェロモン分子であることを様々な側面から検討するとともに、野生ドブネズミのコントロールに応用するための技術開発に取り組む。
