Outline of Research at the Start

本申請課題では、加齢や糖尿病、骨転移巣で、遺伝子発現を制御する転写調節因子ネットワーク機構、エピジェネティクス変化について検討する。時間負荷(高継代数)の作用や生活習慣病による酸化的ストレスの作用を「遺伝子発現をpin-pointで規定するTATA-box直上のCpGメチル化による新規のエピジェネティクス機構」に着目し、個体レベルで検討する。またRANKL産生腫瘍で破骨細胞を誘導する骨巨細胞腫の病理組織や初代培養細胞を用いて、「ヒストン蛋白H3.3活性化による骨吸収メカニズム」を検討し、治療戦略に繋がる知見の集積を目指す。

Outline of Annual Research Achievements

現代の超高齢化社会では筋肉量減少・運動能力低下とともに、性差無く「老人性骨粗鬆症」を来たし、所謂フレイル状態から身体機能や認知機能低下による要介護状態を招くため、医療社会福祉上の重要課題となっている。申請者等は、上記非定型的なエピジェネティクス機構と、加齢現象である①組織や細胞の経年劣化、②糖尿病などの生活習慣病による最終糖化産物(AGE)や酸化的ストレスの蓄積との関連に着目し、加齢や骨芽細胞、マクロファージ培養細胞の高継代数という時間的負荷で、CpGメチル化によるRANKL遺伝子と受容体RANK遺伝子の発現低下を来たすことを明らかにし、学会報告、論文投稿することが出来た。また、糖尿病関連の骨代謝研究の中で、培養骨芽細胞への酸化的ストレス負荷は、骨形成抑制/骨吸収促進に繋がる遺伝子発現profileの変化(p16INK4a増加、RANKL増加、OPG低下、sFRP4増加によるWnt signal低下)を呈することをもとに、sFRP4遺伝子遺伝子改変動物を作成し、このsFRP4が、皮質骨と骨梁組織のバランサーであることを示すことに成功した。さらに酸化的ストレス下および腫瘍との共存下に於ける個体レベルでの骨代謝の変化についても、これまで準備を進めたRANKL可視化GFPマウスおよびTATA-box上流のCpG部位を遺伝子編集でGpGをCpTに変化させ、メチル化を阻害する遺伝子編集マウスを筑波大学との共同で既に作製しており、これまでの報告と合わせ、第113回、日本病理学会春季総会にて宿題報告を行った。現在、論文化が出来ていない研究についてまとめを行い、一流雑誌への投稿に向けてデータ整理、文献整理を進めている。
