Outline of Research at the Start

パーキンソン病の原因は黒質緻密部のドーパミン神経細胞の変性脱落であるが、どのドーパミン神経細胞サブセットの変性脱落がパーキンソン病のどの症状を引き起こすのかはわかっていない。応募者らを含む複数のグループの研究により、黒質緻密部のドーパミン神経細胞は、その領域ごとに不明瞭ながら近接した線条体領域へ投射するトポグラフィーを示すことがわかっているが、この投射様式の違いと遺伝子発現の違いとを結びつける要因はわかっていない。本研究課題では、ドーパミン神経細胞の投射様式と遺伝子発現プロファイリングとの相関を明らかにし、機能的ドーパミン神経細胞サブセットを同定することで、大脳基底核回路の機能的解明を行う。

Outline of Final Research Achievements

In the striatum, direct pathway neurons with the D1R gene and indirect pathway neurons with the D2R gene were found to be localized and projected to specific regions of the substantia nigra pars compacta (Ogata et al., 2022). Recently, it has been reported that adeno-associated virus serotype 1 (AAV1) can efficiently transmit anterograde transsynaptically. However, this technique has been reported to be successful in glutamatergic excitatory and GABAergic inhibitory synapses, but not in dopaminergic synapses. In 2024, we succeeded in the world's first AAV1 anterograde transsynaptic transmission experiment in dopaminergic synapses.

Academic Significance and Societal Importance of the Research Achievements

本課題においては、パーキンソン病の原因とされるドーパミン神経細胞への線条体からの特殊な神経投射を明らかにした(Ogata et al., 2022)。さらに世界で初めてドーパミン作動性シナプスにおけるAAV1順行性経シナプス伝播実験に成功し、より直接的な方法でドーパミン神経細胞の投射先の細胞の同定、その細胞の遺伝子情報および投射様式を同時解析することが可能となった。このことは、ドーパミン神経細胞のサブタイプを遺伝子情報および形態学的、電気生理学的な情報と合わせて、機能的ドーパミン神経細胞サブセットを解明するための大きな一歩となる。
