Outline of Research at the Start

化学遺伝学を用いた経路選択的な操作によって精密な回路操作を行い、課題への影響と神経細胞の活動の変化を調べ、大脳基底核の局所回路の機能同定を行う。特に霊長類大脳基底核の局所回路が、価値・意欲をどのように区別し、どのように不安障害やうつ病が引き起こされるのかを、ヒトと相同な脳構造を持つ霊長類において包括的に明らかにし、ヒトの精神障害の治療につながる神経操作技術の確立に寄与する。

Outline of Annual Research Achievements

霊長類において不安障害に因果的に関わると考えられる神経回路は、大脳皮質－大脳基底核に広く分散して存在すると考えられてきた。近年、応募者らの研究から、このいわゆる「不安回路」の下流域には線条体ストリオソーム構造があることがわかってきた。この線条体ストリオソーム構造は、腹側被蓋野や黒質緻密部 (SNc) にあるドーパミン (DA) 細胞に対する直接投射があることが知られている。化学遺伝学などの遺伝学的手法と、電気刺激法による経路同定を用いて、霊長類の辺縁皮質からストリオソーム構造へ至る神経経路を同定し、ストリオソーム活動の変化が DA 細胞に与える影響を調べる。

Reason

国際共同研究で進めてきたマウスにおけるストリオソーム・マトリックス構造のカルシウムイメージングの研究がNature Commに掲載された。本研究では、非常に多くの数のmedium spiny細胞の記録を、マウスが葛藤課題を遂行中に行い、ストリオソーム・マトリックスの細胞の機能の違いを明らかにすることができた。理論通り学習に関わる信号は、ストリオソームに多いことが分かったが選択的とはいえず、ストリオソームは、幅広い結果に関する情報を表現していることが分かった。本研究の成果をもとに、マカクザルでも細胞種特異的なイメージングの実現を目指す。また、化学遺伝学による経路選択的な操作の基盤となる研究として、NAc-腹側淡蒼球の経路選択的な操作の実験を行っている。予備実験ではあるが、意欲の制御に関わることが明らかになりつつある。以上のことから、これまでの実験だけでなく、現在進行中のものもともに順調に進んでいると判断する。

Strategy for Future Research Activity

化学遺伝学による経路選択的な操作の基盤となる研究として、NAc-腹側淡蒼球の経路選択的な操作の実験を行っている。また、線条体ストリオソームに投射するpACC、cOFCの領野の同定が行えたことから、この領野をターゲットとして、cOFCからストリオソーム系の化学遺伝学による制御へと発展される。
