Outline of Research at the Start

現在までに提案されているラボオンチップの設計手法には,①1:1で液滴を混合する操作のみしか考慮していない,②混合操作などで起こるエラーの影響を考慮していない,③大規模な問題には対応できない,といった問題がある.そのため,近い将来実現すると考えられている,様々な比率での混合操作が可能で,かつ,大規模な実験も可能な次世代のラボオンチップに,既存の設計手法をそのまま適用するのは効率が良くないと考えられる.そこで,本研究構想では,LSI設計の分野における「関数分解」および「タスクスケジューリング」の概念を導入することによって,上記の三点の問題を克服するようなラボオンチップの設計理論の確立を目指す.

Outline of Annual Research Achievements

DMFB(Digital Microfluidic Biochip)の操作時の分割エラーの相殺を利用して,分割エラーによって発生した誤差が目標ノードに与える影響が最小になるように,希釈グラフを変形してエラー訂正を行う手法をすでに開発している.しかし,この手法は目標ノードに近いノードで大規模な分割エラーが発生した際,目標ノードに大きな誤差が生じてしまうという問題点があった.そこで,今年度は,目標ノードに近いノードを二重化し,再希釈を行う手法を検討した.さらに,希釈操作で生成される液滴の出力先のノードを目標ノードからのレベルが等しくなるように希釈グラフを変形することで効率的かつ正確なエラー訂正手法も検討した.これらの新たな手法により,目標ノードににおける平均の濃度の誤差を従来手法に比べて大幅に減少できることを確認した.
Micro Electrode Dot Array(MEDA)で試薬混合を行う場合は,試薬の比率は1:1だけでなく様々な比率での混合が可能である.MEDAにおける3種類以上の試薬からなる目標の液滴を生成する手法として,Division-by-Factor-Method for MEDA (DFMM) が提案されている.しかし,DFMMは複数の目標液滴を生成する場合に液滴を再利用することを考えていない.そこで,中間液滴を再利用することによって,混合に使用する試薬の量を減らすことができる手法を考案した.シミュレーション実験の結果,液滴の再利用によって混合に必要な試薬を削減できることを確認した.その他,MEDAのルーティング手法に関して既存の手法の様々な改良や,一般的なルーティングおよびスケジューリングに関する研究成果を外部発表して,情報処理学会SLDM研究会優秀発表賞,国際会議ICEIC 2024でBest Paper Award Silver Prizeを受賞した.

当初の予定通り操作エラーに対する手法の修正案を考案することができ、また,関連するラボオンチップに関する様々な研究やタスクスケジューリングに関する研究が順調に進展しているため。

Strategy for Future Research Activity

今までと大きな方針は変えずに,次世代の大規模なラボオンチップをエラーも考慮して効率的に使いこなす設計手法を確立するための要素技術の取得を目指して,以下の項目の研究を進める.① 今までに検討を進めたDMFBにおけるエラーの影響を減少できる手法を既存の研究と組み合わせてより良い手法を開発して成果をまとめて外部発表を目指す.② DMFBだけでなくMEDAやPMD向けのエラーの影響を減少させる手法についての検討も行う.③ 今までにMEDAにおける様々な状況における効率的なルーティングやスケジューリング手法,効率的な試薬合成手法の開発を行ってきた.これらを統合して,MEDAにおける効率的な試薬合成を実現するルーティングおよびスケジューリング手法を確立する.また,複雑な状況では最適解を求めることが困難になることが想定されるため,効率的なヒューリィスティックの開発も検討する.④ 今までバイオチップに限らず一般的なチップを対象として,計算資源の使用量を最適に決定するタスクスケジューリング手法を開発している.それらの開発したスケジューリング手法をバイオチップの設計に応用することを検討する.⑤ 上記以外に,ラボオンチップの設計手法に関しての研究動向を調査して,新規の研究テーマを見つけて今後の研究の方向性に関して検討を行う.
