Outline of Research at the Start

磁気マーカーの高調波磁化の粒径dHおよび周波数依存性を実験・数値解析の両面から明らかにする。この結果を基に、粒径dH = 5nmの変化を正確に表す、高精度な高調波磁化ダイナミクスのモデル化を行う。
次に、得られた高調波磁化モデルをベースに、分解能5nmの精度を持った粒径dH推定法を開発する。
最後に、この粒径dH推定という新規な手法を用いた磁気的免疫検査法を開発し、迅速・高感度・高精度なバイオ物質検査システムを実現する。

Outline of Annual Research Achievements

磁気マーカーと結合したバイオ物質を定量検出する磁気的免疫検査は、迅速かつ高感度な免疫検査法として、その開発に大きな期待が寄せられている。しかし、磁気マーカーと検査対象でないタンパク質などとの非特異吸着は、検査精度低下の原因となる。本研究では、結合による流体力学径の変化を、高調波磁化信号を利用して推定するという独自な手法を立案・確立し、検査精度向上の阻害因子を磁気的に識別する手法を開発することを目的としている。本目的を達成するためには,溶液中の磁性ナノ粒子の複雑な磁化ダイナミクスを解明する必要があり、本年度の具体的な研究成果は以下の通りである。
1.磁気マーカーの粒径分布および磁気モーメント分布推定法の開発
目的達成のためには、磁気マーカーの液中粒径分布を推定する必要があるが、高調波磁化信号を用いた提案手法では、粒径分布と磁気マーカーの磁気モーメント分布を同時推定することで、精度が向上することを明らかにした。その同時推定手法を開発し、実験により本手法の有用性を確認しまた。
2.バイオターゲットを用いた磁気的免疫検査実験
上記の手法をもとにした、磁気的免疫検査システムの開発を行った。また、バイオターゲットとしてSARS-CoV-2を用いた免疫検査実験を実施した。今後は、感度向上に向けたシステムの改良を行う予定である。

当初の計画通り、免疫検査装置の開発、バイオターゲットを用いた磁気的免疫検査実験を実施することができた。

Strategy for Future Research Activity

感度向上に向けて、磁気的免疫検査システムの改良を行う。具体的には、励起磁界強度・周波数に適した検出コイルの最適化を行い、ノイズの低減を実現する。次に、改良した装置を用いた磁気的免疫検査実験を行い、本研究の目的である、迅速・高感度・高精度全てを兼ね備えた磁気的免疫検査が可能であることを実証する。
