Outline of Research at the Start

本研究では、過渡回折格子法という、エネルギーや熱力学量や拡散過程を時間分解で観測できる手法の空間分解能と感度を向上させることを目的とする。ヘテロダイン検出法を用いることで、数マイクロメートル程度の空間分解とマイクロ秒程度の時間分解能を併せ持つ検出システムを作る。これを用いて、近年注目されている微小液滴内での拡散運動やタンパク質構造変化を時間分解で観測できるように改造し、バルク中のそれらとどのように異なるかを明らかにする。

Outline of Annual Research Achievements

本研究では、過渡回折格子法の空間分解能と感度を向上させることによって、マイクロメートル程度の空間分解とマイクロ秒程度の時間分解能を併せ持つ検出システムを作る。これを用いて、タンパク質構造変化を時間分解で観測できるように改造し、バルク中のそれらとどのように異なるかを明らかにすることを試みた。
これまでに開発してきたTGシステムをベースにして、微小領域の分子運動や熱力学量計測を可能にするTG測定システムのさらなる改良を行った。新しい対物レンズや非球面レンズを使ったシステムに改造し、これによって、マイクロメートル領域での熱グレーティング信号を検出できた。さらに、空間制限での分子反応ダイナミクスを観測するために、微細加工システムを用いて、マイクロメートルサイズの微小セルを作製し、この作成したセルの中に液体を封入して、TG信号を観測することに成功した。
液滴の性質を明らかにするために、いくつかの報告されているタンパク質を用いて液滴を作成した。また、時間分解で液滴形成を調べるためにTePixDというこれまで我々が発見していた光反応で凝縮が起こるタンパク質の反応を詳細に調べ、その凝縮の原因を追究した。また、多くの研究者が調べているBLUFの光反応に対して、一般的にみられる新しいダイナミクスを発見し、その変化をもたらす分子論的な原因を明らかにすることに成功した。さらに、eBLUFと呼ぶ新しいBLUFタンパク質を見出し、このタンパク質が光状態と暗状態の間でユニークな分子間相互作用を持つことを示した。

Reason

開発してきたTGシステムをベースにして、新しい対物レンズや非球面レンズを使ったシステムに改造し、マイクロメートル領域でのTG信号を観測することに成功した。さらに感度を向上させるため、ヘテロダイン法を用いて、信号を観測することにも成功した。また、よく知られている液滴の性質を観測するために、いくつかの報告されているタンパク質を用いて最適な液滴を作成した。さらに、空間制限での分子反応ダイナミクスを観測するために、微細加工システムを用いて、マイクロメートルサイズの微小セルを作製した。

Strategy for Future Research Activity

さらに小さい領域で高感度な信号の測定が可能なシステムを作成する。一方で、一般に液滴と認識されているタンパク質を用いて液滴を作成し、その熱力学的性質を調べる。また、液滴内での反応と通常の溶液中での反応を比較するために、いくつかのタンパク質で溶液中における反応ダイナミクスを時間分解で観測する。
