Outline of Research at the Start

nmスケールの微小構造体の化学的特性や物性の評価のために高い空間分解能を有する分析法を開発することが喫緊の課題となっている.固体最表面に対しては原子スケールにおける計測法が精力的に開発されている一方,高さ方向に空間分解能を有する計測法は未踏領域として残されている.そこで,本研究では,多岐にわたる化学現象に関与する表面電位に着目し,電極最表面だけでなく,高さ方向にもサブnmオーダーの空間分解能を有する表面電位の三次元計測法を開発する.開発した手法に立脚し,固液界面の電位分布を高分解能で計測し,電気二重層の微視的構造を初めて明らかにする.

Outline of Annual Research Achievements

近年,ナノ科学・テクノロジーへの興味の高まりを背景に,優れた機能を有するnmスケールの微小構造体が盛んに開発されている.これに伴い,微小構造体の化 学的特性,物性の計測を可能とする,高い空間分解能を有する分析法が強く求められている.固体最表面に対しては原子スケールにおける計測法が精力的に開発 されている一方,高さ方向に空間分解能を有する計測法は未踏領域として残されている.そこで,本研究では,多岐にわたる化学現象に関与する表面電位に着目し,電極最表面だけでなく,高さ方向にもサブnmオーダーの空間分解能を有する表面電位の三次元計測法を開発することを目的とする.
開発する手法では,単分子電位計を開発しこれをDNAに結合させることにより表面からの高さを規定し,高さ分解能を実現する.令和3年度においては,足場となるDNAの伝導特性,および電極への結合様式について検討した.令和4年度では,これらの知見に基づき,単分子電位計としてMBを導入したDNA(MB-DNA)の伝導特性を評価した.走査型トンネル顕微鏡を用いた単分子電気伝導計測によりMB-DNAはMB未修飾DNAに比べ伝導度が低下すること,またこれはトンネル障壁の増加に起因することを明らかにした.令和5年度では,電気化学環境下,電極電位に依存してMB-DNAの単分子伝導度が変化することが分かり,伝導度を指標として電位計測が実現できることを明らかにした.

Reason

本研究課題において開発する計測手法においては,単分子電位計と足場DNAを結合させた複合体の伝導特性を指標として,試料表面の面直方向における電位分布を決定する.当該年度においては,MB-DNA複合体の電気伝導度が電極電位に応じて変化することを見出した.さらに,この伝導度変化がMBの
電子状態を介した共鳴伝導に基づき定量的に説明できることが分かった.以上により,提案した原理に基づき電位計測が実現できることが明らかとなったことから,現在までの進捗状況は,当初の計画以上に進展している,と評価する.

Strategy for Future Research Activity

上述の通り,これまでMB-DNAが電極電位を反映した伝導度変化を示し,電位計測が実現可能であることを明らかにした.本手法によって測定可能な電位範囲はDNAと複合化した分子の酸化還元電位近傍であるため,フェロセン等の他の分子についても同様に検討し,電位計測の広いダイナミックレンジを担保する.また,電気化学計測に広く用いられているAu(111)電極に本計測手法を適用し,電極表面における電気二重層の3次元電位分布をnmオーダーの空間分解能にて初めて実験的に明らかにする.
