Outline of Research at the Start

SbzP生成物はNAD類似骨格を持つ。この構造は、NAD結合部に作用して競合阻害する抗腫瘍化合物と類似しており、本化合物についても医薬品活性が期待できる。そこで、下流のアミノアシル基転移反応が進行する様にSbzI, SbzAの酵素反応の基質特異性を改変し、SbzQと共に用いて、アザトリヒドロインダン環を修飾する。天然の基質以外にSbzIに関してはスルホン酸やアミノ酸、SbzAに関しては複数のアミノアシルtRNA合成酵素など非天然型基質の使用を試みる。また、ホモログクラスターより得られた修飾酵素を用いた結晶構造解析とそれに基づく変異酵素反応によって生成物を修飾し、構造多様性の拡大を達成する。

Outline of Annual Research Achievements

SbzPホモログPsePの基質認識機構解明のため、クライオ電子顕微鏡解析を進め、NAD結合構造を2.6Aの分解能で取得した。その結果、NADのアデニンがmonomer BのF457、R466によって、二リン酸がR466によって、ニコチンアミドリボシドがmonomer AのY413、Y418とmonomer BのD462とLoop 6によって保持されることを明らかにした(図c)。また、それぞれのアミノ酸残基に変異導入を行った所、F413A, Y418A, F457A, R466A, Y699Aのいずれも80%以上のアザインダンジヌクレオチド生成の減少が見られ、それぞれの基質保持における重要性が示された。その一方で、アデニン、リン酸の保持に関わるアミノ酸変異体F457A、R466A、Y699AはSAMの β,γ-脱離によって生成するmethylthioadenosine (MTA)の生産量が大きく減少した(>64%)。その一方、F413A、Y418Aでは105%、67%とそれぞれ活性上昇、中程度の減少に止まった。このデータより、それぞれSAM認識における異なる役割が示唆された。また、SAMを酵素構造にドッキングし、結合部位に変異を加え、SAMから合成されるMTAの検出、ストップトフロー解析を行うことによって、SAMの結合能を評価した。また、速度論解析から得られた反応機構を提示し、その矛盾を、東京大学大学院農学生命科学研究科の寺田透教授との共同研究にて、ドッキングシミュレーション、MDシミュレーションによって説明した。カリフォルニア大学Dean Tantillo教授との共同研究にて、反応機構のQM計算を行い、その反応性を精査した。

Reason

SbzPホモログPsePの構造機能解析は完了しつつある一方で、そのホモログ遺伝子の発現や、培養による物質生産の結果、新規なアザインダン化合物を発見、単離するという目的は達成できていない。現在、関連アミノ酸合成酵素や、修飾酵素については、解析を進めているが、その構造解析、反応解析はまだ途中であり、成果報告まで至っていないため。

Strategy for Future Research Activity

単離構造決定や酵素合成に関するポスドクを雇用し、新規物質生産を試みる。様々な培地、培養条件で、菌体培養を行い、新規化合物の取得を試みる。SbzPに関しては、酵素や反応中間体の構造をもとにして、酵素機能改変を行い、新規NAD化合物の合成を試みる。基質が不明なものに関しては、理化学研究所のSPRやITCを適応し、基質と酵素の親和性を評価する。
