Outline of Research at the Start

これまでにタウ病変モデルマウスを用いて2光子顕微鏡による生体脳イメージングを行った。実験結果から、タウ凝集体を有する神経細胞をグリア細胞とマクロファージが貪食して、脳外へと排出するメカニズムが明らかとなってきた。次のステップとして、この老廃物排出メカニズムの全容解明と最終的な機能制御を実現するためには、炎症に伴うグリア細胞やマクロファージの多様な機能変化をとらえる評価系が必要と考えた。そこで細胞内の様々な生理パラメータを計測可能な量子センサ技術を生体脳内のグリア細胞やマクロファージに応用することを考え、そのため技術開発を進めている。最終的にタウ病変マウスの脳細胞への応用を目指す。

Outline of Final Research Achievements

This study identified active waste clearance pathways mediated by meningeal macrophages and brain-resident glial cells that contribute to the progression of brain atrophy in neurodegenerative diseases such as Alzheimer’s disease. In vivo two-photon imaging revealed that microglia and macrophages phagocytose neuronal debris and abnormal proteins and transport them toward the cerebrospinal fluid. Furthermore, we demonstrated that astrocytes contribute to waste clearance via perivascular spaces. These findings provide new insights into the immune-mediated regulation of neurodegeneration.

Academic Significance and Societal Importance of the Research Achievements

アルツハイマー病などの神経変性疾患は、記憶障害や認知機能の低下を引き起こし、要介護状態や寝たきりの原因となることから、超高齢社会において深刻な医療・福祉上の課題となっている。本研究では、これまで不明であった脳内の老廃物除去メカニズムを免疫細胞の能動的関与という視点から解明し、疾患の進行に密接に関与する新たな標的を明らかにした。これにより、認知症の早期診断や進行抑制を目指した革新的な治療法の開発につながる可能性があり、患者の生活の質(QOL)の向上や、介護・医療にかかる社会的コストの軽減に大きく貢献することが期待される。
