This study investigates how the tumor immune environment and suppression change in gynecological cancers (endometrial/ovarian) during disease and treatment to inform new therapies. Prior analysis of patient data (DNA/RNA sequencing) linked molecular features to clinical outcomes. Key findings: cytokine signaling, T cell genes, specific mutations, and gene expression profiles (GEPs) correlated with drug sensitivity and prognosis. Notably, resistance after repeated chemotherapy was separate from immune gene expression. Mouse models of ovarian/endometrial cancers, and immunodeficient mice, tested combining chemotherapy with targeted drugs (VEGF/PARP inhibitors) on the immune landscape. Tumor analysis showed combination therapy boosted CD8+ T cell infiltration more than chemotherapy alone, suggesting a benefit. This research emphasizes the complex interaction between the tumor, immunity, and treatment in gynecological cancers, providing insights for personalized therapies.

| Academic Significance and Societal Importance of the Research Achievements |
