Outline of Research at the Start

我々の体を構成する様々な器官は、その発生過程において厳密に制御され、それぞれの機能に即した特徴的な形態を呈するようになる。器官形成において、それぞれの細胞が無秩序に増殖すると形態形成が阻害されることから、なんらかの細胞間コミュニケーションを行いながら秩序だった器官形成が行われていることが予想される。本研究では、上皮-間葉相互作用において形態が決定されるメカニズムの解明を目指し、新たな器官再生技術の開発に寄与する。

Outline of Final Research Achievements

In this study, we analyzed the gene expression profiles during the early development of organs formed through epithelial-mesenchymal interactions using CAGE and scRNA-seq. And we focused on LYPD1, which is specifically expressed in dental mesenchymal cells. Functional analysis suggested that LYPD1 localizes to lipid rafts in the cell membrane and may play a crucial role in the differentiation process of odontoblasts. Furthermore, this research indicated the potential utility of LYPD1 as a molecular marker for pre-odontoblasts.

Academic Significance and Societal Importance of the Research Achievements

再生医療において、器官再生を成し遂げるには複雑な形成過程の詳細を明らかにし、生体外で再現する技術開発が必要となる。歯は上皮-間葉相互作用により形成される器官として知られており、2種類の細胞が複雑にコミュニケーションを行いながら、器官形成が行われる。本研究により、将来象牙質を形成する前象牙芽細胞のマーカー遺伝子が明らかとなったことで、今後の再生医療への応用が期待される。
