Outline of Research at the Start

日本は高齢化に伴い神経機能の低下による重大事故や神経変性疾患が増加している。しかし,老化にともない低下する神経機能や細胞形態の変容が,神経の変性へと至るプロセスに関する知見は乏しい。申請者は,タンパク質の代謝や異化に関わる酵素,Pin1の欠損マウス(Pin1 KO)を用い加齢に伴う脳の変容を調べてきたところ,視床に特異的にアミロイド凝集体を認め,それらが筋硬直性ジストロフィ患者のエオシン陽性視床封入体と極めて類似した病理学的特徴を有することを見出した.Pin1の老化や神経変性疾患に対する役割を,視床の神経機能を中心に明らかにする。

Outline of Annual Research Achievements

日本は高齢化に伴い神経機能の低下による重大事故や神経変性疾患が増加している。しかし,老化にともない低下する神経機能や細胞形態の変容が,神経の変性へと至るプロセスに関する知見は乏しい。申請者は,タンパク質の代謝や異化に関わる酵素,Pin1の欠損マウス(Pin1 KO)を用い,その行動学,組織学解析を行ってきた。そして,Pin1 KOは,大脳と脊髄および末梢からの情報を統合する重要な領域である視床の神経細胞内にアミロイドが高度に蓄積されることを認めた。当該研究は,①Pin1 KOの表現型はどの神経変性疾患と類似するのか?,②視床のアミロイド封入体は視床の機能に影響するか?,③老化した神経細胞はどのような生理機能変容を有するのか?,④Pin1活性の修飾はどのような神経機能を変えるか?の4つの問いを明らかにし,Pin1の老化や神経変性疾患に対する役割を,視床の神経機能を中心に明らかにする。さらに老化したマウスから神経を単離し,細胞生理機能の加齢変化を直接明らかにする。既年度までにPin1 KOの表現型とヒト神経変性疾患との関連を剖検脳を用いた組織染色により調べてきた.その結果筋硬直性ジストロフィー患者剖検脳に認められるエオシン陽性視床封入体と免疫染色および形態染色の染色性が一致し,極めて性質が類似する封入体であることを認めた.本年度はそれらの封入体の性質を調べるためimmunoblottingにより加齢変化を調べた。野生型マウスとPin1 KOマウスの視床の加齢変化をRIPA可溶性画分と不溶性画分に分け調べたところ,可溶性画分において予想に反して有意な低下を認めている。

Reason

当該年度に代表者が所属が変更となり,施設の立ち上げやセットアップなどの時間を要することとなった.その結果,視床の機能解析に若干の遅れを生じた.

Strategy for Future Research Activity

加齢に伴う視床の封入体に変動が起きているかをRIPA buffer可溶性画分および不溶性画分に分け検討を行っている。興味深いことにPin1 KOの可溶性画分においてアミロイド前駆体の有意な低下を認めており,それらと不溶性画分の関連を調べている。
