Outline of Research at the Start

癌細胞は大量(H+)の酸産生を行うことにより癌細胞膜外超微小ナノスケール空間におけるpHを低下させ、このナノスケール空間におけるpH低下が癌細胞周辺の正常細胞を死滅させ、癌細胞周辺領域に癌細胞増殖スペースを生み出す。食品成分(漢方薬も含む)によって細胞外pH低下を抑制(正常化)することで、癌細胞増殖の抑制が生じ、その結果癌細胞との共存が可能となる。このようなメカニズムにより、特に高齢者にとってのガン発症予防とガン温存療法を試みる。

ヒト食道扁平上皮癌細胞 (Ann Surg Oncol. 27:3218, 2020; Sci Rep 9:16055, 2019; Am J Pathol 189:1973, 2019; Oncotarget 9:29957, 2018; Oncotarget 9:25993, 2018;Oncotarget 9: 23237, 2018; Oncotarget 8:17921,2017; Oncotarget 8:2209, 2017)、ヒト胃癌細胞 (Int J Oncol 55:905, 2019; Gastric Cancer 22:473, 2019; Oncotarget 8:101394, 2017; Cell Physiol Biochem 42:68, 2017)、食道癌幹細胞 (J Gastroenterol 53:197, 2018)、ヒト直腸癌細胞 (J Gastroenterol 50:287, 2015)、ヒト膵癌細胞 (Pancreatol 12:440, 2012)、ヒト肝臓細胞癌細胞 (Int J Oncol 50:1857, 2017; J Cancer 7:1524, 2016)、ヒト乳癌細胞 (Arch Biochem Biophys 539:92, 2013)を用いた。研究代表者の既報の方法(Nature Commun 9:5278, 2018)を用いて、癌細胞周辺の局所空間である超微小ナノスケール空間(細胞膜外近傍である 細胞膜外100 nm 以内の範囲)の細胞外液 pH を測定した。上記癌細胞では、解糖系のみによる糖代謝が行われた結果、大量の酸が生成され、癌細胞周辺超微小ナノスケール空間細胞でのpH低下が確認され、癌細胞が生成する酸性環境により癌細胞周辺正常細胞が障害を受けることが明らかとなった。このことは、癌細胞が酸排泄を介して、癌細胞周囲の正常細胞に障害を与え、正常細胞を破壊することにより、癌細胞の増殖スペースを確保していることを示唆している。

Reason

癌細胞表面のpH測定はおおむね順調に推移している。癌細胞周辺pH低下が周囲の正常細胞に障害を与えることが明らかとなったが、これは癌細胞に比し、正常細胞ではH+排泄イオン輸送体発現が少なく機能面からもH+排泄能が低く、pH低下に対しての抵抗性が低いことが明らかとなった。これらのことから癌細胞が産生したH+が生み出す酸性環境が正常細胞に障害を与えることにより、癌細胞自身のが自らの増殖スペースを確保することが明らかとなった。梅成分であるムメフラールがナトリウムイオン依存的カルボン酸輸送体発現を高めることも明らかとなり、ムメフラールおよびカルボキシル基を含有した食品、いわゆるアルカリ化食品の摂取により、モデル動物の間質液pH改善(上昇)させることも明らかとなった。このことは、これらのアルカリ化食品が間質液pHの緩衝能を増大させれることにより、癌細胞が産生し間質液に排出するH+を緩衝して、pH低下を防ぐ効果を有していることを示し、癌転移を防ぐ可能性を有していることを示唆している。これらのことより、2024年度において、ムメフラールおよびカルボキシル基含有食品摂取により担癌動物モデルにおける癌転移を阻止できるか否かの検証を行うことが可能となったことから、本研究はおおむね順調に進捗するものと考えられる。

Strategy for Future Research Activity

これまでに得られた研究結果により、ムメフラールおよびカルボキシル基含有食品が間質液pH緩衝能を増大させることにより、癌細胞周囲液pH低下(酸性環境形成)を防ぎ、正常細胞への障害を阻止して、癌細胞増殖阻止を行うことができる可能性が示唆された。ムメフラールおよびカルボキシル基含有食品摂取と癌細胞に発現している各種イオン輸送体の阻害剤投与の相乗を検証し、癌細胞の増殖が抑制されるか否かを検証するとともに癌転移の程度も2024年度において検証する。さらに、2024年度においてはムメフラール以外にナトリウムイオン依存性カルボキシル基輸送体発現増大をもたらす食品成分の探索も試みる。このような研究を推進することにより、特に体力低下を来している高齢者における癌温存療法開発の試みを行う。
