Outline of Research at the Start

本研究は、生体筋に近似した3D骨格筋培養細胞をプラットフォームとし、マイオカイン分泌に好適な運動・栄養処方を模倣する生理的刺激を検出するとともに、それら処方(運動・栄養処方)をヒトへ応用展開する、統合的アンチエイジングのための生体応用研究基盤を創成することを目的とする。
当該研究基盤創成の結果、運動・栄養処方に対する認知機能、運動機能、体組成の変化などの詳細かつ多面的なヒト生体応用検討が可能となる。
運動・栄養処方を基軸としたQOL向上や健康寿命延伸のための研究拠点形成が期待できる。

Outline of Annual Research Achievements

骨格筋の減退化が健康寿命阻害の初発段階であることを鑑みると、健康寿命の延伸に効果的な処方を策定するにあたり、運動や外的刺激がもたらす骨格筋の機能向上機序を細胞レベルで把握し、それが他の器官にどういった好適な作用を誘発し、ひいては生体システムを好適化するか、生体という複雑系に応用展開する実践的研究基盤形成が必要である。この需要に訴求すべく、本研究は、生体筋に近似した3D骨格筋培養細胞をプラットフォームとし、マイオカイン分泌に好適な運動・栄養処方を模倣する生理的刺激を検出するとともに、それら処方(運動・栄養処方)をヒトへ応用展開する、統合的アンチエイジングのための生体応用研究基盤を創成することを目的としたものである。
今年度は、3D骨格筋培養システムを活用し、筋肥大効果を有することが明らかとなったマカ(栄養素)と電気刺激による筋収縮(運動)の併用効果の検証を実施した。マカと電気刺激の併用によって、効果的に筋肥大を惹起することを認め、国際誌に公表した。さらに、3D培養筋の収縮システムに関する論文も公表した。
加えて、統合的アンチエイジングのためのヒトへの実践的応用(運動・栄養処方)方略の効果検証も実施した。一過性の運動・栄養(必須アミノ酸)処方により、認知機能の亢進効果を明らかにした。この成果を、国際誌に公表した。さらに、高強度間欠的運動が愛情ホルモンとも言われるオキシトシンの分泌に及ぼす影響、異なる運動様式が、一種のマイオカインに位置付けられる乳酸産生に及ぼす影響、カフェイン摂取がジャンプパフォーマンスに及ぼす影響について、国際誌に公表した。
このように、培養細胞並びにヒトを対象とした研究を往還して進めている。

Reason

3D骨格筋培養システムを活用し、一定の成果を挙げ、国際誌にて公表されたことが理由である。3D培養筋の収縮システムに関する論文ならびにマカ(栄養素)と電気刺激による筋収縮(運動)の併用効果の検証に関する論文である。
また、一過性の運動と栄養の認知機能の亢進に対する併用効果についても、ヒトを対象に実験を実施することができ、有意義な結果を得て、国際誌にて公表されたことも理由である。さらに、カフェインの介入によるジャンプパフォーマンス向上効果についても有意義な知見を得、国際誌にて公表された。加えて、長時間の低酸素環境が認知パフォーマンスに及ぼす影響、高強度間欠的運動がオキシトシン分泌や脳循環制御に及ぼす影響など、国際共同研究成果も得られた。

Strategy for Future Research Activity

引き続き、運動に栄養を組み合わせることで、効果的に認知実行機能や筋機能を改善し得る方略を検討していく。解析中のデータや知見は整理し、まとめて、学術論文にて公表する。また、投稿中の論文についても学術論文にて公表する。さらに、健常若年男性に対する一過性のレジスタンス運動と栄養素の併用効果の検証実験結果についても、論文にまとめていく。その際、栄養素とマイオカイン分泌、そして認知機能の関係について、詳細に検討する。
このように、培養細胞並びにヒトを対象とした研究を往還して進めていく。
