Outline of Research at the Start

放射線はヒト体内にあるDNAに対して様々な形状の損傷を与える。その多様なDNA損傷の中で、DNA二重鎖が同時に切断されるDNA二本鎖切断は最も重篤なDNA損傷の一つとされている。一方で人体は、傷ついたDNAを復元する「DNA修復」という機能を持つことから、正確にDNAを修復できるかどうかがその後の人体の運命決定に大きな影響を与える。本研究ではDNA修復の精度に関わる最重要分子である53BP1を対象に、その詳細な分子機構解明に挑戦する。人体がどのようにして正確なDNA修復を実現するか、その仕組みを明らかにすることで、放射線障害の低減に貢献することができると考えている。

Outline of Annual Research Achievements

放射線照射が誘発するDNA二本鎖切断(DSB)は重篤なDNA損傷の一つとされ、その修復の成否は細胞の運命決定に大きな影響を与える。ヒト細胞に生じたDSBは、非相同未端連結(NHEJ)または相同組換え(HR)のいずれかによって修復される。我々は、放射線照射直後、DSB近傍ではNHEJに適したコンパクトなクロマチン構造が構築され、HRの進行と伴にクロマチン構造が変化することで、NHEJからHRへの切り替えを促進していることを明らかにしてきた。そのクロマチン環境変化を制御する因子は53BP1であることが示唆されているが、その詳細な分子機構は未解明である。本研究では、細胞がどのようにしてクロマチン環境をNHEJからHRに適した構造へと変化させているのか、DSB発生から時間の経過と伴に局在を変化させる53BP1 repositioning(再配置)」と呼ばれる現象に着目し、その分子機構の解明に迫る。昨年度、超高解像度蛍光顕微鏡3D-SIM(OMX)により53BP1を観察した結果、53BP1は複数のナノドメインを構成していることが明らかになった。我々が所有する53BP1変異体にて53BP1ナノドメインを解析した結果、特に53BP1-olimerization変異体にて53BP1ナノドメイン構築に異常が認められた。次に、環境変化が53BP1の集積に影響を及ぼすかどうか検討するため、細胞内のATP濃度が低下した条件下において53BP1などのDSB修復因子の集積を解析した。その結果、53BP1の集積低下、およびRAD51形成能の低下が認められた。

超高解像度蛍光顕微鏡3D-SIM(OMX)により53BP1は複数のナノドメインの可視化に成功し、その機能解析が進んでいる。

Strategy for Future Research Activity

昨年度までにゲノムワイドな部位特異的DSB誘導系を用いたChIP-seq/ATAC-seq解析のための条件検討を重ねてきた。そのため本年度は、DSB近傍のクロマチン構造と53BP1ナノドメイン機能の関係性についてさらに解析を進める。
