Outline of Research at the Start

本研究の目的は,酸性条件下で高い活性を持つ酸性ウレアーゼと食品廃棄物の骨粉に着目し,酸性から中性のpH領域で尿素分解によるアンモ
ニア生成を抑制しながらリン酸カルシウム化合物(CPC)を析出させ,地盤材料を固化させる環境配慮型のバイオセメンテーション技術を新た
に開発することである。
本研究の独創性と新規性は,自然界で生物が作るバイオミネラルをセメント物質として用いること,骨粉をリン酸カルシウム源として有効利
用すること,アンモニアが発生しない環境配慮型のバイオセメンテーションであること,地盤工学,環境工学,コンクリート工学,微生物学,
地質学,鉱物学,地球化学などの学際的研究であること,などである。

Outline of Annual Research Achievements

2年目となる令和5年度は,①微生物由来の酸性ウレアーゼの探索,②培養試験およびリン酸カルシウム化合物(CPC)室内析出試験,③小型容器を用いた地盤材料の室内固化試験,を実施した。その結果,得られた主な成果の概要は,次のとおりである。
①微生物由来の酸性ウレアーゼの探索:単離源として植物や動物を対象とし,ウレアーゼを有する乳酸菌の単離試験を異なる培養条件下で実施した。その結果,好気条件よりも嫌気条件の方が多くの乳酸菌を単離することができたが,いずれもウレアーゼ活性が非常に低いことがわかった。一方,製品評価技術基盤機構(NITE)のNBRC(NITE Biological Resource Center)から購入した乳酸菌3株のウレアーゼ活性を調べた結果,高いウレアーゼ活性を有する乳酸菌が1株あることがわかった。また,同菌株のウレアーゼ活性のpH依存性について調査した結果,pH3~4の酸性領域で高いウレアーゼ活性を示した。
②培養試験およびリン酸カルシウム化合物(CPC)室内析出試験:前記した乳酸菌株を用いた温度25~37℃の培養試験では,温度が菌体濃度に及ぼす影響は小さく,いずれも24時間後の菌体濃度が最大となる。ウレアーゼ活性については,温度が高いほど活性が高くなった。遠沈管を用いたCPC析出試験に関しては,CPC源として食品廃棄物である動物(牛と豚)の骨粉とマグロの骨が使用可能であること,析出した結晶の鉱物名はBrushiteであること,などがわかった。
③小型容器を用いた地盤材料の室内固化試験:骨粉およびマグロの骨をCPC源とし,シリンジを用いた砂質地盤材料の室内固化試験を実施した。その結果,固化した供試体の針貫入試験によって得られた推定一軸圧縮強さ(推定UCS)は,温度37℃で試験期間7日の場合に供試体の上,中,下部において,それぞれ37MPa,34MPa,8MPaとなった。

令和5年度における研究の目的,実施計画,経費などの策定が適切であったこと,また,期待された研究成果が概ね順調に得られたこと,さらに,研究代表者に健康上の問題が発生しなかったことなどが,主な理由として考えられる。

Strategy for Future Research Activity

本研究課題の最終年度となる令和6年度においては,①リン酸カルシウム化合物(CPC)室内析出試験,②小型容器を用いた地盤材料の室内固化試験,③提案する技術の有効性評価,について実施し,報告書を作成して本研究課題を終了する。
