Outline of Research at the Start

研究の目的欄にも記載したように、本研究では「ヒドロキシルアミン化学の新展開」と題し、ヒドロキシルアミンの持つ可能性をより深く、かつ多角的に探究し、その秘められた反応性を明らかにする。具体的には以下の4つの課題を重点的に実施する。
1) デザイン型ヒドロキシルアミンを用いた高選択的求電子的アミノ化によるα-アミノ酸類縁体の立体網羅的合成
2) 求電子的アミノ化を利用するヘテロ芳香環から擬ヘテロ芳香環への骨格編集
3) ヒドロキシルアミンを用いた高選択的求電子的エーテル化
4) 炭素―炭素単結合との触媒的シグマボンドメタセシス

Outline of Annual Research Achievements

本年度はまず、前年度の懸案であったα-アミノ酸類縁体の立体網羅的合成に取り組んだ。残念ながら期待した立体網羅性の発現には至らなかったものの、立体選択性の制御因子を見出しつつある。前年度に偶然見出していたトリフルオロメチルアルケンの化学が大きな展開を見せ、これをトリフルオロメチル”チオ”アルケンの化学へと拡張することに成功し、成果を速報として国際誌に掲載した。より一般的なトリフルオロメチルカルコゲンの化学を確立すべく、現在更なる検討を続けている。
当初計画にあった「求電子的アミノ化を利用するヘテロ芳香環から擬ヘテロ芳香環への骨格編集」の課題については難航したが、現在別視点からの骨格編集技術を見出すに至っており、想定していた方法論とは異なるものの、目標とする分子変換を達成しつつある。一方で、上記課題が難航したため、次年度以降予定していた「ヒドロキシルアミンを用いた高選択的求電子的エーテル化」と「炭素―炭素単結合との触媒的シグマボンドメタセシス」の一部を前倒しで検討した。その結果、双方とも順調な進展が見られた。求電子的エーテル化の化学は汎用性の高い新たな求電子的酸素化剤のデザイン、創成に至っており、含酸素分子の新たな立体選択的触媒合成手法を拓きつつある。炭素―炭素単結合メタセシスにおいても、ニッケルの特異な触媒機能を見出すことに成功し、こちらは現在成果を国際誌に投稿、審査中である。
以上のように、課題全体としては概ね順調な進展が見られている。

Reason

研究実績の概要欄にも記載した通り、一部の課題が難航しているものの、前倒しで検討した計画は順調に進んでいる。また、当初予定していなかったトリフルオロメチルカルコゲンの新しい合成化学を拓くことに成功しており、すでに成果発表にも至っている。難航している課題も、別視点からのアプローチによって解決策が見つかりつつあり、当初の研究計画を達成するに十分な手応えがある。以上のように、課題全体としては概ね順調な進展が見られていると判断できる。

Strategy for Future Research Activity

現時点で難航している「α-アミノ酸類縁体の立体網羅的合成」と「求電子的アミノ化を利用するヘテロ芳香環から擬ヘテロ芳香環への骨格編集」に関しては今後も鋭意検討を続けるが、現在大きな展開をみせつつある課題にも積極的に取り組みたい。具体的には「トリフルオロメチルカルコゲンの合成化学」「ヒドロキシルアミンを用いた高選択的求電子的エーテル化」「炭素―炭素単結合との触媒的シグマボンドメタセシス」の三つである。いずれも、当初計画と同等ないしはそれ以上の波及効果を当該分野にもたらす内容である。前者二つに関しては、特に立体選択的な触媒開発に向けて、後者はより広範かつ一般性の高い炭素―炭素単結合の利用へ向けて、展開を図りたい。
