Outline of Research at the Start

MALDI-MS技術によるプロテインタイピング技術は抗体やオリゴヌクレオチド等のプローブ不要の生物分類技術であり、生命科学研究のゲームチェンジ技術の一つとして社会へ浸透している。一方、癌抗原、胎児抗原、血清型等、実用分子指標である糖鎖構造情報取得はイオン化効率の問題により取り残されている。本課題は代表者が開発した糖鎖選択的イオン化技術を鍵としたMALDIグリコタイピン技術基盤を構築するものである。

Outline of Annual Research Achievements

昨年度までに構築したグリコタイピングのさらなる対象拡大と実証研究を実施した。具体的には糖鎖選択的イオン化法を活用した微生物O抗原解析技術、O-結合型(ムチン型)糖ペプチド構造解析技術、および糖鎖捕捉技術glycoblottingと弱イオン交換技術を高度に融合した硫酸化およびリン酸化糖鎖の迅速識別分析技術についてそれぞれ検体を拡大し、実証試験および問題点の抽出と克服を実施した。グラム陰性菌O抗原解析ではジデオキシヘキソースを有する分解しやすい糖残基を有する抗原多糖に着目した解析を行い、ジデオキシヘキソース(colitose)を2残基有する大腸菌O111血清型の解析において、市販のO111LPSおよび、ナショナルバイオリソースを通じて入手した腸管出血性大腸菌O111株のMALDIグリコタイピングを行ったところ、それぞれ報告された構造と異なるO111多糖の異性体が存在することを見だした。そこで、未知O111異性体のNMR解析を実施し、colitose分岐部のglucoseがGlcNAcに置換された構造を有することを同定した。仮性結核菌O抗原を対象とした解析では、デオキシヘキソース含有O抗原特徴的配列パターンと配列同定制度を高める新規マトリックス組成をそれぞれ発見した。また、不糖糖タンパク質の分解挙動についてO-結合型糖鎖に加え、ペプチド側の断片化パターンを含めたMALDI-TOF/TOF MS解析により、特に環状ペプチドにおいてペプチド鎖のアミド結合と側鎖トレオニン残基双方において特徴的な断片化パターンを見だした。硫酸化糖鎖解析ではヒト血清を対象とした大規模グライコミクスを実施し、乳がん患者において、硫酸化糖鎖、フコシル化硫酸化糖鎖、シアリル化硫酸化糖鎖いずれも顕著に増加すること、これらの変化がステージ1の段階で顕著に生じることを見だし、早期診断マーカーとなる可能性を示した。

Reason

当初計画では新規分析対象の開拓を主な標的としていたが、さらに、癌早期診断マーカーの発見や微生物抗原同定法としての改良、新発見O抗原の精密構造同定、糖ペプチド内部構造解析の新規パターン情報など当初目標を大幅に上回る解析結果を得ることにそれぞれ成功した。

Strategy for Future Research Activity

過去3年間に蓄積したMALDIグリコタイピング技術の大規模検証を推進し、次世代プロジェクトへの基板構築を実施する。特に、微生物O抗原解析では大腸菌と赤痢菌を中心とした大規模解析を実施し、新しい臨床診断技術としての可能性を検証する。
