Outline of Research at the Start

本研究では、放線菌シグナル分子の誘導因子としての分子基盤の解明および汎用的二次代謝覚醒手法の開発を行う。まず、ブテノライド型シグナル分子生合成について、遺伝子破壊や標識化合物などを用いて明らかにする。次いで、SRB 受容体ホモログの相同性解析や遺伝子破壊を行い、ブテノライド型分子の認識残基などの構造情報や二次代謝誘導との相関性を解明する。そして高い相同性を有する放線菌ライブラリーから優先的に「シグナル分子 SRB の添加による休眠二次代謝覚醒」に取り組み、新規ゲノムマイニング手法の確立を目指す。

Outline of Annual Research Achievements

本研究課題では、放線菌シグナル分子の誘導因子としての分子基盤の解明および汎用的二次代謝覚醒手法の開発を行う。まず、ブテノライド型シグナル分子生合成について、遺伝子破壊や標識化合物などを用いて明らかにする。次いで、SRB 受容体ホモログの相同性解析や遺伝子破壊を行い、ブテノライド型分子の認識残基などの構造情報や二次代謝誘導との相関性を解明する。そして高い相同性を有する放線菌ライブラリーから優先的に「シグナル分子 SRB の添加による休眠二次代謝覚醒」に取り組み、新規ゲノムマイニング手法の確立を目指す。2023年度は以下の課題を中心に展開した。
「シグナル分子制御系の分子基盤の解明(受容体の共通性・相違性)」
110株の放線菌株におけるブテノライド型シグナル分子の検出を目指し、抽出物を調製してStreptomyces rocheiのシグナル分子合成欠損株に添加実験を行ったところ、いずれの抽出物でも抗生物質の誘導生産を引き起こさなかった。このことは、ブテノライド型シグナル分子の構造特異性を強く示唆している。
「SRB型シグナル分子による休眠二次代謝遺伝子の発現誘導」
放線菌にはシグナル分子合成酵素よりも過剰に受容体ホモログが存在しており、他のシグナル分子を認識する可能性が示唆された。本課題では、二次代謝誘導における SRB の汎用性を見いだすべく、各種単離株に対してSRB を添加し、非添加サンプルとの比較代謝プロファイル解析を行った。薄層クロマトグラフィー(TLC)および高速液体クロマトグラフィー (HPLC)で比較解析したところ、105株中27株において代謝変動を確認出来た。SRBにより誘導生産された二次代謝産物は、通常非生産である可能性が高いため、その構造・活性新規性が大いに期待できる。また、抗マラリア活性試験やミズクラゲの誘導制御因子の探索を行い、非誘導菌株でも顕著な生理活性物質を見いだした。

Reason

当該年度に実施した2課題のうち、まず「シグナル分子制御系の分子基盤の解明」に関し、110株の放線菌株におけるブテノライド型シグナル分子の検出を試みた。S. rocheiのシグナル分子合成欠損株に対して供試菌の培養抽出液を添加したところ、いずれも抗生物質生産の回復は認められなかった。このことはブテノライド型シグナル分子の構造特異性を示す成果と言える。
また、「SRB型シグナル分子による休眠二次代謝遺伝子の発現誘導」に関して、上述の110菌株に対して、ブテノライド型シグナル分子を添加したところ、30%程度の菌株において代謝産物プロファイル変化が見いだされた。このことは、シグナル分子による代謝誘導が可能であることを示唆しており、さらに各放線菌株のリセプターが構造の異なるシグナル分子を寛容に認識しうることも示している。HPLC, TLCによる代謝プロファイルの変化解析に加え、抗マラリア活性試験やミズクラゲの形態分化誘導制御物質の探索を行った。

Strategy for Future Research Activity

今後は、上述の研究課題をより推進すべく、まず「SRB型シグナル分子による休眠二次代謝遺伝子の発現誘導」に関して、放線菌分離源として、新たにインドネシア・フィリピンとの共同研究株を視野に入れている。さらに今後は様々な生物活性試験に供し、生理活性を有する二次代謝産物の代謝誘導を目指す。具体的には、抗菌活性、抗真菌活性、抗マラリア活性、抗トリパノソーマ活性、ミズクラゲ・ブラインシュリンプなどの水圏生物の形態分化誘導・阻害活性の検出を指標にして、新規生理活性物質の獲得を目指す。
また、顕著な代謝産物プロファイル変化が見いだせた株に関しては、大量培養とシグナル分子添加実験を組み合わせ、誘導生産された化合物の構造決定を目指す。
