Outline of Research at the Start

養豚業に大きな損害を及ぼす豚熱(CSF)ウイルスと非定型豚ペスチ(APP)ウイルスは、妊娠豚に持続感染し、胎子の流産や新生子豚の先天性神経異常症を引き起こすと考えられているが、両ウイルスの病原性の分子基盤には不明な点が多い。この研究計画では、CSFウイルスおよびAPPウイルスの病原性、特に持続感染を引き起こす分子基盤を解明するために、細胞の内・外からその機能が発揮できるウイルス構造蛋白質Ernsに着目する。ウイルスがブタに持続感染し、結果として胎子や新生子豚に病原性を発揮するきっかけとなる宿主免疫からの回避と制御された細胞死の抑制に関するErns蛋白質の機能を調べる。

Outline of Final Research Achievements

To elucidate the molecular basis of persistent infection caused by classical swine fever (CSF) virus and atypical porcine pestivirus (APP) virus, we focused on the structural protein Erns, which must be involved as a pathogenic factor in both viruses. We investigated the function of the Erns protein in immune evasion and suppression of regulated cell death. As a result, we revealed that the N-terminal region of the CSF virus is crucial for persistent infection in pigs. Furthermore, this region was involved in the suppression of innate immunity and necroptosis. A similar mechanism was predicted for the APP virus; however, the wild-type virus showed minimal proliferation in piglets, preventing further elucidation of its details.

Academic Significance and Societal Importance of the Research Achievements

CSFウイルスやAPPウイルスの持続感染においてErnsタンパク質のN末端領域が宿主免疫の回避やネクロトーシス抑制に関与していることは、ウイルス-宿主相互作用の理解を深める重要な発見となった。一方、APPウイルスの研究において子豚での増殖が限定的だった点は、ウイルスの自然界における生態に関する新たな研究課題の提案につながると考えられる。また今回の成果は、効果的なワクチンの開発や持続感染を抑制する新たな治療法の開発に貢献できる基盤的な成果と考えられる。
