Outline of Research at the Start

FlhAはべん毛蛋白質輸送の選別輸送センターかつ輸送エンジンとして働く。これまでに、分子選別機能が異常になったflhA変異体やエネルギー共役のしくみが著しく変化したflhA変異体を数多く単離した。本研究では、FlhAの動的機能構造基盤を原子レベルで解明するため、変異型FlhAの構造変化を高分解能で解析できるX線結晶構造解析法、働く現場での蛋白質輸送過程に伴うFlhAの構造変化を活写できるクライオ電子線トモグラフィー法、および変異体の遺伝学的・生化学的機能解析をうまく融合させることにより、FlhAが実際に働いている現場での動的機能構造解析を行う。
