Outline of Research at the Start

ミトコンドリアは、細胞が必要とするエネルギーを作る非常に重要な場所である。従来の研究は、ミトコンドリアがどのようにして作られてくるかに着目していたが、本研究は、機能が悪くなったミトコンドリアを取り除く(分解する)現象であるマイトファジーに着目し、その生理機能を明らかにしようとするものである。具体的には、マイトファジーの誘導が抑制された培養細胞やマウスを用いて、どのような異常が生じるのかを観察する。

Outline of Final Research Achievements

We analyzed BNIP3/NIX_DKO cells as receptor-mediated mitophagy-deficient cells and found that receptor- mediated mitophagy protects cells from ferroptosis by maintaining low levels of mtROS. Furthermore, we revealed that the role of the mitophagy receptors BNIP3 and NIX in mitophagy is to mediate tight attachment and expansion of the isolation membrane to mitochondria.

Academic Significance and Societal Importance of the Research Achievements

マイトファジーは、異常もしくは不要となったミトコンドリアを選択的に分解することでミトコンドリアの機能維持に貢献しており、ミトコンドリア機能低下を伴う様々な疾患の病態や老化現象に関与していると考えられている。このため、マイトファジーを医療へ活用しようとする研究が進められている。本研究は、レセプター依存的マイトファジーの生理的意義や分子機構の一端の解明に成功しており、今後のマイトファジーの医療応用への基盤となる。
