| Outline of Research at the Start |

| Outline of Final Research Achievements |
This study aimed to elucidate the mechanisms governing the integration and morphogenesis of skeletal muscle, cartilage/bone, and tendon in the musculoskeletal system. We focused on identifying muscle-tendon junction-inducing factors, uncovering the role of long non-coding RNAs (lncRNAs) in tendon-bone boundary formation, and elucidating mechanisms of tendon progenitor cell induction. Single-nucleus RNA sequencing (snRNA-seq) revealed several candidate factors potentially involved in muscle-tendon interactions. Knockout analysis of a novel lncRNA indicated its involvement in cartilage differentiation and the morphogenesis of the tendon-bone interface. In addition, we identified direct downstream targets of the tendon-specific transcription factor Scleraxis (Scx) and carried out functional studies. These findings provide new insights into the role of tendon cells in musculoskeletal morphogenesis.
