Outline of Research at the Start

近年、多様な動物の脳において光受容タンパク質様遺伝子が発現していることが報告され、動物の脳における未知の光利用の可能性が注目されている。特に、一般に「光受容能はない」とされている哺乳類の脳において光受容タンパク質様遺伝子が発現しているという知見は興味深い。しかしながら、哺乳類の脳で発現が認められる光受容タンパク質様遺伝子が、実際に光受容体として機能するという明確な証拠がないため、哺乳類の脳内光受容能の真偽は依然として不明である。そこで本研究では、哺哺乳類の脳内光受容タンパク質様遺伝子についてタンパク質レベルの機能解析を行い、それを起点として哺乳類の脳内光受容の解明を目指す。

Outline of Annual Research Achievements

近年、多様な動物の脳において光受容タンパク質様遺伝子が発現していることが報告され、動物の脳における未知の光利用の可能性が注目されている。特に、一般に「光受容能はない」とされている哺乳類の脳において光受容タンパク質様遺伝子が発現しているという発見は、この定説を覆す可能性があり大きな意味を持つ。しかしながら、哺乳類の脳で発現が認められる光受容タンパク質様遺伝子が、実際に光受容体として機能するという明確な証拠がないため、哺乳類の脳内光受容能の真偽は依然として不明である。そこで本研究では、哺乳類の脳内光受容の解明を目指し、哺乳類の脳内光受容タンパク質様遺伝子についてタンパク質レベルの機能解析を起点とした研究を展開している。その結果、2023年度は以下の研究成果を得た。
・光受容体として機能することを明らかにした哺乳類(哺乳類X)の脳内光受容タンパク質Opn3について、駆動するシグナル伝達系を明らかにする目的で、昨年度までに構築した分割型ルシフェラーゼによるGタンパク質活性化の解析系(NanoBiT-Gタンパク質解離アッセイ)を用いて、共役するGタンパク質の同定を試みた。その結果、哺乳類XのOpn3は、全16種のGタンパク質αサブユニットいずれについても光依存的な活性化を示さなかった。
・哺乳類Xの脳内光受容タンパク質Opn3について、single cell transcriptome解析を行い、脳の特定の部位における発現およびOpn3発現細胞の遺伝子発現プロファイルを得た。

Reason

昨年度に構築した実験系を用いて、予定通りの解析が実施出来た。また、組織学的解析においては、当初計画していた手法(in situ hybridization)はうまくいかなかったが、別の手法(single cell transcriptome)により、予想以上の成果が得られた。

Strategy for Future Research Activity

光受容能が証明された哺乳類XのOpn3を中心に解析を進め、今年度明らかにできなかったOpn3が駆動するシグナル伝達系の解明および、哺乳類XにおけるOpn3発現細胞の遺伝子発現プロファイルが得られたので、その情報をもとに、Opn3の生理・分子機能の解析を進める。
また、他の哺乳類の脳内光受容タンパク質の光受容能についても引き続き検討する。
