Outline of Research at the Start

近年、多様な動物の脳において光受容タンパク質様遺伝子が発現していることが報告され、動物の脳における未知の光利用の可能性が注目されている。特に、一般に「光受容能はない」とされている哺乳類の脳において光受容タンパク質様遺伝子が発現しているという知見は興味深い。しかしながら、哺乳類の脳で発現が認められる光受容タンパク質様遺伝子が、実際に光受容体として機能するという明確な証拠がないため、哺乳類の脳内光受容能の真偽は依然として不明である。そこで本研究では、哺哺乳類の脳内光受容タンパク質様遺伝子についてタンパク質レベルの機能解析を行い、それを起点として哺乳類の脳内光受容の解明を目指す。

Outline of Annual Research Achievements

近年、多様な動物の脳において光受容タンパク質様遺伝子が発現していることが報告され、動物の脳における未知の光利用の可能性が注目されている。特に、一般に「光受容能はない」とされている哺乳類の脳において光受容タンパク質様遺伝子が発現しているという発見は、この定説を覆す可能性があり大きな意味を持つ。しかしながら、哺乳類の脳で発現が認められる光受容タンパク質様遺伝子が、実際に光受容体として機能するという明確な証拠がないため、哺乳類の脳内光受容能の真偽は依然として不明である。そこで本研究では、哺乳類の脳内光受容の解明を目指し、哺乳類の脳内光受容タンパク質様遺伝子についてタンパク質レベルの機能解析を起点とした研究を展開している。その結果、2022年度は以下の研究成果を得た。
・従来の研究で用いられていたヒト、マウス以外に13種の哺乳類について脳内光受容タンパク質の同定および培養細胞発現系を用いた組換えタンパク質の解析を行った。その結果、一種の哺乳類の脳内光受容体について、実際に光受容タンパク質として機能する可能性を強く示唆するデータを得た。
・哺乳類の脳内光受容タンパク質が駆動するシグナル伝達系を明らかにする目的で、共役するGタンパク質を同定するための解析系の導入を進め、哺乳類がもつ全16種のGタンパク質αサブユニットについて、分割型ルシフェラーゼを用いたアッセイ（NanoBiT-Gタンパク質解離アッセイ）の実験系を構築した。

Reason

初年度に哺乳類の脳内光受容タンパク質の光受容能を示唆するデータが得られたことは予想以上の進展である。その他、基盤となる実験系の構築も順調である。

Strategy for Future Research Activity

光受容能が示唆された哺乳類の脳内光受容タンパク質について解析を進め、光受容能を完全に証明するとともに、駆動するシグナル伝達系などの光受容タンパク質としての詳細な性質を明らかにする。また、他の哺乳類の脳内光受容タンパク質の光受容能についても引き続き検討する。
