Outline of Research at the Start

神経シナプスでは、情報を伝える側のシナプス前部と、情報を受け取る側のシナプス後部が協調して機能することが不可欠である。近年の超解像観察により、シナプス前部とシナプス後部のシナプス伝達に関わる蛋白質は、ナノメートルレベルで対面整列していることが分かってきた。本研究では、(1)シナプス前部、後部の蛋白質が対面整列を維持する分子機構を明らかにし、(2)疾患において、この機構がどのように破綻するのかを明らかにする。

Outline of Annual Research Achievements

近年の超解像観察により、シナプス前部の神経伝達物質放出に関わるタンパク質とシナプス後部の受容体集積に関わるタンパク質は、シナプス膜局所でナノドメインを形成しており、これらがナノスケールで対面整列することが精緻なシナプス伝達に重要であることが分かってきた(ナノカラム仮説)。また、神経細胞-グリア細胞間においても、特殊なナノスケールの膜ドメインが報告され始めた。本研究では、(1)ナノドメインやナノカラムの形成に液-液相分離が関与しているか、(2)構成タンパク質の疾患変異がナノカラム形成と相分離に与える影響を明らかにする。さらに、(3)神経細胞-グリア細胞間の相互作用における相分離の関与と機能を明らかにすることを目的とした。
2024年度は、前年度に引き続き、シナプスタンパク質によるナノドメイン形成やナノカラム形成における液-液相分離の役割について解析を進めた。具体的には液-液相分離能を選択的に欠失した変異を見出し、多価結合の重要性を明らかにした。さらに、その変異体を用いて、シナプスナノ構築に与える影響を細胞、脳組織レベルで調べる実験系を確立して、検証実験を行った。また、個体レベルの影響を調べるための実験系の構築を進めた。また、神経細胞軸索とオリゴデンドロサイト相互作用によって形成される特殊な膜区画について、前年度までに見出したLGI3-ADAM22/23の機能 (Miyazaki et al. Cell Rep 2024)に着目して、特に複合体の結合様式について研究を進めた。

Reason

1.シナプス-ナノドメイン構築に関わるタンパク質群に関して、液-液相分離能を選択的に欠失した変異体の作製に成功し、液-液相分離の生理機能を調べる実験系を確立した。
2.前年度までに見出した傍パラノードの形成機構に重要なタンパク質複合体の結合様式を明らかにした。

Strategy for Future Research Activity

1.液-液相分離能を選択的に欠失した変異体のシナプスナノ構築に与える影響を細胞、組織、固体レベルで明らかにする。
2.傍パラノードの形成機構に重要なタンパク質複合体の結合様式を構造学的に調べる。
