Outline of Research at the Start

血中・尿中キラルアミノ酸の網羅的定量により、非侵襲・低侵襲での腎不全の早期診断、原因疾患鑑別マーカーを探索する。腎不全は慢性型の患者が多く、早期診断が予防と治療に直結する。応募者は予備試験で、キラルを識別した正確なアミノ酸定量により腎不全の鋭敏なマーカーが発見でき、血中・尿中の含量比較により原因疾患の鑑別が可能であるという結果を得た。そこで本研究では多次元HPLCにより網羅的定量法を開発し、多数の臨床検体を用いた含量解析を行うと共に動物モデルによる機序解析を行う。本研究の達成は侵襲性の低い血液・尿分析による腎不全の早期診断、原因疾患鑑別を可能とし、人々の健康寿命延長に貢献する。

Outline of Final Research Achievements

Simultaneous analytical methods and rapid analytical methods for chiral amino acids have been developed, and a novel biomarkers for kidney disfunction were screened. Evaluation of their amounts in the healthy volunteers was also carried out. For the determination of chiral amino acids, three-dimensional and four dimensional LC systems were used, and an analytical platform for all proteinogenic amino acids were designed. For the 3D-LC system, a reversed-phase, an anion-exchange and an enantioselective columns were used, and a reversed-phase and an enantioselective columns plus tandem mass spectrometry were used for the 4D system. By the analysis of plasma/urine samples of chronic kidney disease patients, D-glutamic acid and D-phenylalanine were found in addition to D-alanine, D-asparagine, D-proline and D-serine. Their amounts are low in healthy volunteers, indicating that these D-amino acids are the useful biomarker candidates for early diagnosis of kidney disfunction.

Academic Significance and Societal Importance of the Research Achievements

血中・尿中キラルアミノ酸定量により非・低侵襲での腎不全の早期診断マーカーを探索した。腎不全は慢性型の患者が多く、早期診断が予防と治療に直結する。本研究では逆相、イオン交換、光学分割による三次元LC、逆相、イオン交換とタンデム質量分析を組み合わせた四次元LCを構築してタンパク質構成全キラルアミノ酸定量の基盤技術を開発した。臨床検体を用いてバイオマーカーを探索し、従来報告されているD-アミノ酸に加えて、D-グルタミン酸、D-フェニルアラニンなど健常人では殆ど存在しない鋭敏なマーカー候補を発見した。本研究の達成は侵襲性の低い血液・尿分析による腎不全の早期診断を可能とし、人々の健康寿命延長に貢献する。
