Outline of Research at the Start

天然資源から強力なアセチルコリンエステラーゼ(AChE)阻害活性を示すフペルジンAやアルツハイマー治療薬ガランタミン等が単離されてきた。これらの化合物には,神経成長因子促進作用を有することも報告されている。AChE阻害作用に基づく薬剤は,脳内AChEのバランスを整えることで記憶障害の改善を図る対象治療薬となるが,その誘導体に神経成長因子促進作用の増強を見い出すことができれば,神経変異疾患に対する根治治療薬の開発に繋がることが大いに期待される。本研究では,フペルジンAとガランタミンの生合成に関わる酵素に変異を導入することで,これらの類縁体生産に資する機能を改変した酵素を生み出すことを試みる。

Outline of Annual Research Achievements

AChE阻害作用に基づく薬剤は,脳内AChEのバランスを整えることで記憶障害の改善を図る対象治療薬となるが,その誘導体に神経成長因子促進作用の増強を見い出すことができれば,アルツハイマー病等の神経変異疾患に対する根治治療薬の開発に繋がることが大いに期待される。昨年度において,ガランタミン等のアマリリスアルカロイドの生合成に関わる3種の4-O-メチル化酵素(4OMT)と基質アナログである3,4-ジヒドロキシベンズアルデヒド(1)の複合体結晶構造解析に成功し,1の4位と3位水酸基へのメチル化効率を制御している機構を明らかにした。今年度は,基質であるノルベラジンとの複合体結晶構造を取得し,この解析をもとに230番目のアスパラギン酸に変異を導入することで,ノルベラジン3位水酸基のメチル化に特化した4OMT機能改変酵素の創出に成功した。本機能改変酵素を用いることで,シュード型アマリリスアルカロイドの効率的生産が可能になることが期待される。また,今年度は,アマリリスアルカロイドの生合成において,ノルベラジンへの変換を担うノルベラジン合成酵素(NBS)と還元酵素(NR)の1のアナログへの基質特異性について検討することで,その酵素反応には3位水酸基が必須であること,一方,その3位メチル化体はNBSの基質にはならないことなどを明らかにした。これにより,一般的なアマリリスアルカロイドの生合成は,ノルベラジンの4位水酸基のメチル化を経て生合成される可能性が高いことを明らかにした。また,シュード型アマリリスアルカロイドの生合成は,ノルベラジンの3位水酸基のメチル化を経て生合成される可能性とともに,未知のNBSホモログとNRホモログが関与することでその生合成がなされることを提案するに至った。

Reason

当初の計画通りに,4OMTの基質との複合体結晶構造の取得と機能改変酵素の創出,及びNBSとNRのアルデヒド基質に対する基質特異性を明らかにすることができたことから,総合的に概ね計画通りに進展していると考えている。

Strategy for Future Research Activity

NBSとNRの基質アナログへの基質特異性についてさらに検討する。また,本年度において得られた4OMT機能改変酵素とNBS及びNRを組み合わせることで新規化合物群の創出を目指す。さらに,4OMT以降の生合成に関与する生合成酵素についても触媒機構の解明と機能改変に取り組むことで,さらなる新規化合物群の創出を試みる。
