Outline of Research at the Start

Outline of Final Research Achievements
This study demonstrates how nanvuranlat, a selective inhibitor of the amino acid transporter LAT1, regulates cell cycle progression in cancer cells. In pancreatic cancer cells, LAT1 inhibition reduced intracellular amino acid levels, leading to activation of the MKK3/6-p38α MAPK pathway. This triggered phosphorylation and proteasomal degradation of cyclin D1, a key regulator of G1 phase, thereby blocking the G1/S transition. The same molecular response was observed in mouse xenograft tumors, confirming the in vivo relevance of the MKK3/6-p38α-cyclin D1 axis. These findings collectively suggest the presence of a previously uncharacterized cell cycle checkpoint that monitors amino acid availability and modulates proliferation accordingly. This newly identified amino acid-dependent checkpoint adds a critical layer to our understanding of nutrient sensing in cancer and provides mechanistic insight into the antiproliferative effect of LAT1-targeted therapies.

Academic Significance and Societal Importance of the Research Achievements
